摘要
为满足小麦高分子量谷蛋白亚基(HMW-GS)高通量分子检测的需求,以小麦种子为材料,以传统CTAB法为基础,结合磁珠法,创新了一种操作简便、成本低廉、周期短的小麦种子高通量DNA提取新方法,并采用紫外分光光度计、琼脂糖凝胶电泳、AS-PCR等方法验证了DNA的产率和质量。结果显示,采用新方法提取的基因组DNA产率和质量稳定,满足了小麦HMW-GS分子检测的要求。
To meet the demand of high-throughput molecular detection of HMW-GS in wheat, based on the traditional CTAB method and magnetic bead method, this study created a new method of high-throughput DNA extraction of wheat seeds, which was simple operation, low cost, short cycle. The yield and quality of DNA were verified by ultraviolet spectrophotometer, agarose gel electrophoresis and AS-PCR. The results showed that the yield and quality of the genomic DNA extracted by the new method were stable,and met the requirement of wheat HMW-GS molecular detection.
