铁皮石斛多糖的闪式提取工艺及质量评价 被引量:9

摘要 目的建立铁皮石斛多糖的闪式提取工艺及质量标准。方法采用闪式提取器常温快速提取铁皮石斛粗多糖,正交试验优化液料比、提取次数、提取温度等参数。对24批不同产地、不同品种石斛的多糖糖含量及单糖组成进行聚类分析,拟定铁皮石斛多糖质量标准。结果铁皮石斛多糖闪式提取最佳工艺条件为:液料比30m L·g^(-1),浸泡30 min后,常温提取3次,每次提取2 min。采用优化工艺制备的铁皮石斛多糖的糖含量为70.68%~99.82%,单糖组成中甘露糖与葡萄糖比例为3.75~25.60,可与水草石斛、金钗石斛多糖明确区分。结论闪式提取铁皮石斛多糖工艺高效、稳定,所制备的粗多糖糖含量不低于70%,甘露糖与葡萄糖的含量比值不低于3.5。 Objective To establish the optimum process of homogenate extraction and quality standard of polysaccharides from Dendrobium officinale.Methods Polysaccharides were obtained by homogenate extractor rapidly at room temperature.Liquid-solid ratio,extracting temperature and times were selected as independent variables of orthogonal test to determine the optimal process.The quality standard of Dendrobium officinale polysaccharides were determined by clustering analysis on contents and monosaccharide compositions of polysaccharides from 24 batches of Dendrobium officinale,Dendrobium aphyllum and Dendrobium nobile from different habitats. Results The optimum parameters were as follows: liquid-solid ratio 30 m L·g-1,room temperature,extract for 3 times,2 min each time,after 30 min soaking.The content of the polysaccharides extracted from Dendrobium officinale by the optimal process was 70.68% 99.82% and the ratio of mannose to glucose was 3.75 25.60,which were quite different from those of Dendrobium aphyllum and Dendrobium nobile.Conclusion The homogenate extraction process of polysaccharides from Dendrobium officinale was stable and efficient. The purity of the prepared Dendrobium officinale polysaccharides was not less than 70%,and the ratio of mannose to glucose was not less than3.5.
