京大戟多糖的提取及苯酚-硫酸法测定其多糖含量的研究 被引量:9

摘要 对京大戟多糖的提取条件和多糖的测定方法进行探索,以期优化京大戟多糖的提取条件和测定方法。采用热水浸提法提取京大戟多糖,同时采用苯酚-硫酸法测定其多糖含量。结果显示:(1)京大戟多糖最佳提取条件为液料比30∶1、提取次数为2、提取温度为80℃、提取时间为2 h;(2)苯酚-硫酸法测定京大戟多糖含量的最佳条件为:苯酚浓度为5%、浓硫酸用量为5 m L、反应温度为100℃、显色时间为30 min,在此条件下,测得京大戟中含糖量为22.09%。苯酚-硫酸法的精密度和重现性的RSD为1.38%、稳定性RSD为1.78%、加标回收率为98.35%,表明该实验获得的提取条件和测定方法均可有效用于京大戟多糖的提取和测定。 To explore the extraction conditions of polysaccharides from Euphorbia pekinensis and the conditions for the determination of polysaccharides, in order to select the better extraction conditions and the better conditions for the determination of the polysaccharide method of the Euphorbia pekinensis. The polysaccharides extracted from Euphorbia pekinensis are extracted by- hot water and the content of polysaccharides is determined by phenol sulfuric acid method. The polysaccharides of Euphorbia pekinensis in this condition under the liquid material ratio 30: 1, the number of extractions is 2 times, the extraction temperature is 80 ℃ , and the extraction time is 2 h. The conditions for the extraction are as follows : the concentra- tion of phenol is 5% , the concentration of concentrated sulfuric acid is 5 mL, the reaction temperature is 100℃ , and the color time is 30 min, and the polysaccharide content in the Euphorbia pekinonis is determined as the content 22.09% , the precision and repeatability of phenol sulfuric acid method are RSD, 1.38% , stability RSD 1.78% , and standard addition recovery98. 35%. The extraction conditions and determination methods can be used as a feasible method for the extraction and determination of polysaccharides in Euphorbia pekinensis
