摘要
目的:探讨中藏药黄芪对CCl_4致小鼠急性肝损伤的保护作用研究,为保肝药物研究及中藏药的开发利用提供试验基础.方法:以50%乙醇和水分别进行提取得到浸膏.小鼠随机分组,即正常对照组、CCl_4模型组、联苯双酯阳性组及各药物乙醇取得物和水提取物的高、中、低剂量组.各给药组给药7d后,CCl_4制备小鼠急性肝损伤模型.测定血清丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)、丙二醛(MDA)的含量及小鼠肝脏指数的变化.结果:黄芪醇提物与水提物低、中、高三个剂量组均可降低CCL致小鼠肝损伤模型中MDA、ALT、AST水平,黄芪醇提物高浓度与水提物高浓度可显著降低小鼠MDA(P<0.05),醇提物低、中与水提物低、中浓度极显著降低MDA水平(P<0.01),黄芪水提物较醇提物作用更显著.黄芪水提物低浓度能显著降低ALT水平(P<0.05),其余各组均极显著降低ALT、AST水平(P<0.01).
Aim:The protective effects of Astragalus on acute liver injury induced by CCl4 in mice were studied in this paper to provide a basis for the study of hepatoclitic drug and the development and utilization of medium Tibetan medicine.Methods:Extract the extract with 50% ethanol and water respectively.The mice were randomly divided into a normal control group,CCl4 model group,biphenyl diester positive group,and the high,medium and low dose groups of ethanol obtained and water extract of each drug.The acute hepatic injury model of mice was prepared by CCl4 after each drug group was given 7 d.Determination of serum alanine aminotransferase(ALT),aspartate aminotransferase(AST),malondialdehyde(MDA)content and changes in mouse liver index.Results:Astragalus alcohol extract and water extract from low,medium and high three dose groups all can reduce CCl induced liver injury in mice model of MDA,ALT,AST levels,Astragalus alcohol extraction of high concentration and the water extract from high concentration can significantly decrease the MDA in mice(P〈0.05),alcohol extraction of low,medium and low water extract,concentration significantly decrease the MDA level(P〈0.01),Astragalus water extraction of the alcohol extract more prominent role.Low concentration of Astragalus water extract significantly decreased ALT level(P〈0.05),while the other groups significantly reduced ALT and AST levels(P〈0.01).
