通窍活血汤对血管性痴呆大鼠学习记忆能力及海马突触可塑性影响 被引量:10

Effects of Tongqiao Huoxue Decoction on Learning and Memory Ability and Hippocampal Synaptic Plasticity of Vascular Dementia Model Rats

摘要 目的:研究通窍活血汤对血管性痴呆(Vascular dementia,Va D)大鼠学习记忆能力及海马突触可塑性的影响。方法:选取40只清洁级健康雄性SD大鼠,随机数字表法分为正常组、模型组、中药组、安慰剂组,采用Olsson法建立Va D模型,造模成功后,中药组予以通窍活血汤(1 m L/100 g·d)灌胃4周,安慰剂组予蒸馏水灌胃4周。Morris水迷宫实验测试其学习记忆能力,TUNEL法检测海马CA1区神经元凋亡情况,在体海马CA1区场电位实验检测大鼠海马长时程增强(LTP),观察神经元突触可塑性的变化。结果:中药组Va D大鼠平均逃避潜伏期较安慰剂组明显缩短,穿越平台次数明显增多,海马CA1区神经元凋亡比率显著下降(P〈0.05),海马高频刺激(HFS)后1 h末场兴奋性突触后电位(f EPSP)的平均斜率保持在(138.61%±3.79%),明显高于安慰剂组的(112.72%±2.63%)(P〈0.05)。结论:通窍活血汤能减少神经元丢失率,改善神经可塑性和突触重构,改善Va D大鼠学习记忆能力。 Objective:To study the effects of Tongqiao Huoxue Decoction to the learning and memory ability and the hippocampus synaptic plasticity of the vascular dementia(Va D)rats. Methods:40 clean healthy male SD rats were randomly divided into normal group,model group,TCM group and the placebo group. Established Va D model by Olsson method. After the model was successful,the rats in TCM group were fed Tongqiao Huoxue Decoction(1 m L·100 g^-1·d^-1)for 4 weeks,the placebo group rats were fed distilled water for 4 weeks. Morris water maze tested their learning and memory abilities. TUNEL assayed hippocampal CA1 neurons apoptosis. LTP in hippocampus was induced to detect the synaptic plasticity changes. Results:The neuronal apoptosis in the CA1 region of the hippocampus of TCM group was significantly lower than the placebo group,the learning and memory impairment was significantly enhanced,and the difference was statistically significant(P〈0.05). The average slope of the f EPSP after HPS 1 hour in the hippocampus of TCM group was(138.61%±3.79%),the placebo group was(112.72%±2.63%),and the difference was statistically significant(P〈0.05). Conclusion:Tongqiao Huoxue Decoction can decrease the neuronal apoptosis in hippocampal CA1,improve the neural plasticity and synaptic remodeling,and then improve learning and memory impairment in Va D rats.
