摘要
目的:研究羟基红花黄色素-a与芍药苷对大鼠脑缺血再灌注损伤((CIRI)的保护作用及其对PI3K和AKT蛋白表达的影响。方法:取SPF级雄性SD大鼠均参与建模,建模成功后用随机数字法将其分为羟基红花黄色素-a组、羟基红花黄色素-a加芍药苷组、芍药苷组、氯吡格雷组和模型组(每组20只);采用大鼠大脑中动脉线栓法复制局灶性脑缺血模型,建模成功后羟基红花黄色素-a组、羟基红花黄色素-a加芍药苷组、芍药苷组、氯吡格雷组和模型组大鼠,均给予相关的方法治疗;采用Westernblotting方法测定各组大鼠PI3K和AKT蛋白水平的含量,比较各组用药后的疗效。结果:羟基红花黄色素-a加芍药苷组大鼠用药后神经功能缺损评分值均低于羟基红花黄色素-a组、芍药苷组、氯吡格雷组和模型组(P<0.05);羟基红花黄色素-a加芍药苷组大鼠脑梗死面积均小于羟基红花黄色素-a组、芍药苷组、氯吡格雷组和模型组(P<0.05);羟基红花黄色素-a加芍药苷组大鼠PI3K、Akt蛋白基因水平均低于羟基红花黄色素-a组、芍药苷组、氯吡格雷组和模型组(P<0.05)。结论:羟基红花黄色素-a加芍药苷用于抗脑缺血再灌注损伤有助于提高其临床疗效,能通过抑制PI3K和AKT蛋白水平实现脑组织的保护作用。
Objective:To study the protection of hydroxysafflor yellow-a combined with paeoniflorin on cerebral ischemia reperfusion injury(CIRI)rats and its influence on expression of pi3k and akt protein.Methods:All SPF male Sprague Dawley rats were involved in modeling.After successful modeling,the rats were randomly divided into hydroxysafflor yellow-a group,hydroxysafflor yellow-a and paeoniflorin group,paeoniflorin group,clopidogrel group and model group,20 in each group.All rats were treated by a middle cerebral artery occlusion method to replicate the focal cerebral ischemia model.After successful modeling,the hydroxysafflor yellow-a group,hydroxysafflor yellow-a and paeoniflorin group,paeoniflorin group,clopidogrel group and model group were all treated by relevant methods;PDGF levels in cerebrospinal fluid and serum were detected by enzyme-linked immunosorbent assay;PDGF-mediated PI3KAKT signaling pathway was determined in each group by Western blotting.The clinical effects of each group were compared.Results:The scores of neurological deficits after treatment with hydroxysafflor yellow-a+PDGF beta-receptor antagonist group were lower than those of hydroxysafflor yellow-a group,PDGF beta-receptor antagonist group,clopidogrel group and model group(P〈0.05);saffron yellow-a+PDGF beta-receptor antagonist group cerebral infarct size,was less than that of hydroxysafflor yellow-a group,PDGF beta-receptor antagonist group,clopidogrel group and model group(P〈0.05);The levels of PI3K and Akt protein genes in safflower a+PDGFβ-receptor antagonist group were lower than those in hydroxysafflor yellow-a group,PDGFβ-receptor antagonist group,clopidogrel group and model group(P〈0.05).Conclusion:Hydroxysafflower yellow-a and paeoniflorin were used in the treatment of cerebral ischemia and reperfusion injury to improve its clinical efficacy,and could achieve the protective effect of brain tissue by inhibiting the level of PI3K and AKT protein.
