机械张力加载时间对成纤维细胞机械传导效应影响

摘要 目的探讨加载机械张力参数为拉伸幅度的10%时,机械张力的加载时间对正常皮肤成纤维细胞(NSFB)机械传导效应的影响。方法体外培养人体NSFB和增生性瘢痕成纤维细胞(HSFB),NSFB和HSFB分别设实验组和对照组,实验组加载机械张力参数为拉伸幅度10%,对照组不加载机械张力。在加载周期性机械张力3、5、7d时,用CCK-8检测各组细胞增殖能力,用Real-time PCR法检测各组细胞Integrin-β_1、P130Cas基因表达水平,用Western-Blot方法检测细胞Integrin-β_1、P130Cas蛋白表达水平。结果加载机械张力5d时,NSFB实验组与HSFB对照组比较,细胞增殖、P130Cas和Integrin-β_1的表达水平差异均无统计学意义(Z=-0.218,F=0.591、0.079,P>0.05);加载3、7d时,NSFB实验组与HSFB对照组比较,细胞增殖、P130Cas、Integrin-β_1表达差异均有统计学意义(Z=-5.294^-2.411,F=34.182~872.148,P<0.05)。结论拉伸幅度为10%的机械张力加载5d时,可使NSFB表现出与HSFB相同的机械传导效应。 Objective To investigate the effect of time under mechanical tension(TUMT)on the mechanotransduction of normal skin fibroblasts(NSFB)when mechanical tension was set to 10%stretch amplitude. Methods Human NSFB and hypertrophic scar fibroblasts(HSFB)were cultured in vitro as test group and control group,respectively.The test group was loaded with mechanical tension of 10% stretch amplitude,while the control group was loaded with zero tension.When cyclic TUMT reached 3,5,and 7 d,cell proliferation in each group was determined by CCK-8 assay,while the mRNA and protein expression of Integrin-β1 and P130 Cas in each group were measured by real-time PCR and Western blot,respectively. Results When TUMT was 5 d,there were no significant differences between the test group and the control group in cell proliferation and the expression of Integrin-β1 and P130 Cas(Z=-0.218,F=0.591 and 0.079,P〉0.05);but when TUMT was 3 or 7 d,there were significant differences between the test group and the control group in cell proliferation and the expression of Integrin-β1 and P130 Cas(Z=-5.294 to-2.411,F=34.182-872.148,P〈0.05). Conclusion When TUMT of 10%stretch amplitude is 5 d,NSFB may exhibit the same mechanotransduction as HSFB.
