益智提取物对大鼠肝纤维化的防治作用 被引量:1

Prevention and treatment of hepatic fibrosis in rats by Alpinia oxyphylla extract

摘要 目的探讨益智乙酸乙酯提取物对大鼠肝纤维化防治作用及其作用机制。方法采用高脂饲料喂养12周诱导SD大鼠非酒精性脂肪性肝炎模型,实验分为对照组、模型组、益智乙酸乙酯提取物组(实验组)。实验组给予高脂饲料喂养+四氯化碳(CCL4)腹腔内注射,每天给予益智提取物500 mg/kg灌胃;模型组给予高脂饲料喂养+CCL4腹腔内注射,每天给予相同体积的白开水灌胃;对照组,正常饲料喂养,每天给予相同体积的白开水灌胃,持续4周。外周血检测甘油三酯(TG)、总胆固醇(TC)、谷草转氨酶(AST)、谷丙转氨酶(ALT);ELISA检测肝组织透明质酸(HA)、层粘连蛋白(LN)含量。肝组织检测丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH)、超氧物歧化酶(SOD)活性。RT-PCR检测转化生长因子-β1(TGF-β1)m-RNA、Smad2、Smad3、Smad7表达。Masson染色观察肝纤维化情况。结果与对照组比较,模型组的TG、TC、AST、ALT、HA、LN、MDA、Smad2、Smad3、TGF-β1 m-RNA水平明显升高,差异有统计学意义(P<0.05);GSH、SOD、Smad7明显下降,差异有统计学意义(P<0.05)。与模型组比较,实验组的TG、TC、AST、ALT、HA、LN、MDA、Smad2、Smad3、TGF-β1 m-RNA水平明显下降,差异有统计学意义(P<0.05),GSH、SOD、Smad7明显上升,差异有统计学意义(P<0.05)。Masson染色下,与对照组比较,模型组及实验组肝脏组织均出现纤维化。与模型组比较,实验组肝组织脂肪变性情况改善,但肝纤维化情况改善不明显。结论益智乙酸乙酯提取物能抗氧化应激、下调TGF-β1/Smad信号通路,有缓解肝纤维化的作用。 Objective To investigate the effect and mechanism of ethyl acetate extract of Alpinia oxyphylla on theprevention and treatment of hepatic fibrosis in rats.Methods SD rats were induced by high-fat diet for 12 weeks.The ratswere divided into control group,model group,and ethyl acetate extract group(experimental group).The experimentalgroup was given high fat diet and carbon tetrachloride(CCL4)intraperitoneal injection,and every day treated with Alpinia oxyphylla extracts 500 mg/kg gavage for 4 weeks;model group was given high fat diet and CCL4 intraperitoneal injectionevery day,given the same volume of white boiling water gavage for 4 weeks;control group was given normal diet every day,given the same volume of white boiling water by gavage for 4 weeks.Triglyceride(TG),total cholesterol(TC),aspartatetransaminase(AST)and alanine aminotransferase(ALT)were detected in peripheral blood,and hyaluronic acid(HA)andlaminin(LN)contents in liver tissue were detected by ELISA.The activities of malonic dialdehyde(MDA),glutathione(GSH)and superoxide dismutase(SOD)were detected in the liver tissue.The expression of transforming growth factor-β1(TGF-β1)m-RNA,Smad2,Smad3 and Smad7 was detected by RT-PCR.The liver fibrosis was observed by Massonstaining.Results Compared with the control group,the levels of TG,TC,AST,ALT,HA,LN,MDA,Smad2,Smad3 and TGF-β1 m-RNA in the model group were significantly increased while GSH,SOD and Smad7 were significantlydecreased,the differences were statistically significant(P〈0.05).Compared with the model group,the levels of TG,TC,AST,ALT,HA,LN,MDA,Smad2,Smad3 and TGF-β1 m-RNA in the experimental group were significantly lower thanthose in the model group while GSH,SOD and Smad7 were significantly higher,the differences were statistically significant(P〈0.05).For Masson staining,compared with the control group,the liver tissues of the model group and the experimentalgroup all appeared fibrosis.Compared with the model group,the fatty degenerat
