摘要
目的探讨阿托伐他汀对急性心肌梗死(acute myocardial infarction,AMI)小鼠缺血心肌的影响及可能机制。方法 30只SPF级C57小鼠随机分为AMI组、阿托伐他汀治疗组和假手术组,每组10只;AMI组、阿托伐他汀治疗组结扎心脏左前降支建立AMI模型,假手术组仅行开胸、不结扎左前降支;术后24h,阿托伐他汀治疗组给予阿托伐他汀10mg/kg灌胃,AMI组、假手术组给予等量生理盐水灌胃,均1次/d,共7d。术后第8天,取各组小鼠外周血检测血清同型半胱氨酸(homocysteine,Hcy)、白细胞介素-6(interleukin-6,IL-6)、肌酸激酶同工酶(creatine kinase-MB,CK-MB)、心肌肌钙蛋白T(cardiac troponin T,cTnT)水平,并进行比较。结果 AMI组小鼠血清CK-MB[(404.66±27.05)u/L]、cTnT[(148.78±8.54)ng/L]、Hcy[(17.68±2.39)μmol/L]、IL-6[(284.54±37.84)pg/L]水平高于假手术组[(344.22±34.94)u/L、(128.85±4.38)ng/L、(9.22±1.38)μmol/L、(165.78±21.64)pg/L]和阿托伐他汀治疗组[(353.71±35.17)u/L、(134.72±7.46)ng/L、(13.78±1.51)μmol/L、(188.66±13.29)pg/L](P<0.05),阿托伐他汀治疗组与假手术组比较差异无统计学意义(P>0.05)。结论阿托伐他汀对AMI小鼠心肌有保护作用,其机制可能是通过抑制IL-6、Hcy表达而抑制心室重塑、炎性反应和AMI发展,减轻心肌损伤。
Objective To observe the effect of atorvastatin on acute myocardial infarction (AMI) in mice and its possible mechanism. Methods Thirty SPF mice were divided into AMI group, atorvastatin group and sham operation group, with 10 mice in each group. The left anterior descending branch was ligated to establish AMI models in AMI group and atorvastatin group, and was not ligated in sham operation group. Atorvastatin group received lavage with 10 mg/kg atorvastatin, and the other two groups received lavage with the equivalent volume of normal saline once a day totally for 7 d. At the postoperative 8 day, the levels of homocysteine (Hcy), interleukin-6 (IL-6), creatine kinase-MB (CK-MB) and cardiac troponin T (cTnT) in the peripheral blood were detected and compared among groups. Results The serum levels of CK-MB ((404.66±27.05) u/L), cTnT ((148. 78±8. 54) ng/L), Hcy ((17. 68±2. 39) μmol/L) and IL-6 ((284.54±37. 84) pg/L) in AMI group were significantly higher than those in sham operation group ((344. 22 ± 34.94) u/L, (128. 85±4. 38) ng/L, (9. 22± 1. 38) μmol/L, (165. 78±21. 64) pg/L) and atorvastatin group ((353.71μ35.17) u/L, (134.72±7.46) ng/L, (13.78±1.51) t±mol/L, (188.66± 13.29) pg/L), and there were no significant differences between sham operation group and atorvastatin group (P〉0.05). Conclusion Atorvastatin has a protective effect on AMI mice probably by inhibiting ventricular remodeling, inflammatory reaction and the development of AMI to alleviate cardiac injury by reducing the expressions of IL-6 and Hcy.
