摘要
目的研究大孔吸药剂科附树脂纯化新疆芜菁总黄酮的工艺条件。方法以比吸附量、吸附率、解吸率为指标,采用静态吸附对5种大孔吸附树脂进行筛选;采用UV法以上样液浓度、吸附流速、树脂柱径高比、泄露曲线、洗脱剂浓度、吸附次数、洗脱曲线等参数为指标对纯化工艺进行筛选。结果用HPD-100大孔树脂纯化新疆芜菁总黄酮的最佳工艺为:层析柱径高比1∶10,上样速率2mL/min,上样质量浓度0.24mg/mL,重复吸附1次,洗脱剂95%乙醇,洗脱剂用量1BV。结论 HPD-100树脂适合于新疆芜菁总黄酮的分离与纯化。
Objective To study the purification technology of total flavonaoids of Xinjiang Brassica rapaL.Methods 5 different macroporous resins were compared by static absorption method with adsorption capacity,adsorption rate,desorption rate as indexes.Using the concentration of sample solution,flow velocity,ratio of diameter to height,adsorption-leakage curve,concentration of eluent,times of adsorption,elution curve as indexes,the purification technology was optimized by UV.Results The optimal purification technology of HPD-100 macroporous resin was as follows:the rate of diameter to height is 1:10,sampling flow rate as 2 mL/min,the concentration of the sample solution flow rate was 0.24 mg/mL,repeated adsorbing again,eluting with 95% ethanol,elute volume was 1 BV.Conclusion The HPD-100 macroporous resin can be applied to separate and purify of total flavonoids of the Xinjiang Brassica rapaL.
