摘要
目的:探讨LMP1、COX-2与鼻咽癌恶性生物学行为的相关性。方法:选取本院收治的鼻咽癌患者86例,取鼻咽癌组织及癌旁组织标本,采用western-blot法测定其中LMP1、COX-2蛋白表达量,采用荧光定量PCR法测定其中增殖、侵袭基因mRNA的表达量,进一步采用Pearson检验评估鼻咽癌组织中LMP1、COX-2蛋白表达量与增殖、侵袭基因表达量的相关关系。结果:鼻咽癌组织中LMP1、COX-2蛋白的表达量均显著高于癌旁组织;增殖基因c-Myc、Dub3、Sam68mRNA的表达量高于癌旁组织;侵袭基因NONO、Glypican-3、DEPDC1、TRIM27 mRNA的表达量高于癌旁组织,PKIP mRNA的表达量低于癌旁组织(P<0.05)。鼻咽癌组织中LMP1、COX-2蛋白表达量与增殖、侵袭基因mRNA的表达量直接相关。结论:鼻咽癌组织中存在LMP1、COX-2蛋白高表达,且具体表达量与肿瘤恶性生物学行为直接相关。
Objective: To study the correlation between LMP1, COX-2 and malignant biological behavior of nasopharyngeal carcinoma. Methods: A total of 86 cases of nasopharyngeal carcinoma patients in our hospital during January 2015 to February 2017 were chosen as study subject, nasopharyngeal carcinoma tissues and paracancerous tissues were token and expression of LMP1, COX-2 protein in them were measured by Western-blot method. Expression of proliferation and invasion genes mRNA were measured by fluorescence quantitative PCR method, further correlation between LMP1, COX-2 and malignant biological behavior was evaluated by Pearson test. Results: The expression of LMP1 and COX-2 protein in nasopharyngeal carcinoma were significantly higher than those in paracancerous tissue; expression of proliferation genes such as c-Myc, Dub3, Sam68 mRNA were higher than those in paracancerous tissue; expression of invasion genes such as NONO, Glypican-3, DEPDC1, TRIM27 mRNA were higher than those in paracancerous tissue, expression of PKIP mRNA was lower than that in paracancerous tissue( P 〈0.05). The expression of LMP1 and COX-2 protein in nasopharyngeal carcinoma were directly related to the expression of proliferation and invasion gene mRNA. Conclusion: There is high expression of LMP1 and COX-2 protein in nasopharyngeal carcinoma tissues, and the specific expression is directly related to the malignant biological behavior of tumor.
