摘要
目的:探讨加味黄连温胆汤对慢性肾衰竭(CRF)模型大鼠脑组织MDA、SOD、Na^+-K^+-ATP酶表达的影响。方法:将40只Wistar大鼠随机分为空白对照组、假手术组、CRF模型组、中药组,每组10只。空白对照组、假手术组、CRF模型组予以生理盐水灌服,中药组予加味黄连温胆汤灌胃。6周后检测各组大鼠脑组织MDA、SOD、Na^+-K^+-ATP酶水平。结果:与空白对照组、假手术组比较,CRF模型组、中药组MDA含量均明显升高(P〈0.05);与CRF模型组比较,中药组MDA含量明显降低(P〈0.05);与空白对照组、假手术组比较,CRF模型组SOD含量均明显降低(P〈0.05),中药组无明显差异;与CRF模型组比较,中药组SOD水平明显升高(P〈0.05);与空白对照组、假手术组比较,CRF模型组、中药组Na^+-K^+-ATP酶活性均明显降低(P〈0.05);与CRF模型组比较,中药组Na^+-K^+-ATP酶活性明显升高(P〈0.05)。结论:加味黄连温胆汤可显著降低5/6肾切除大鼠脑组织MDA含量、提高SOD、Na^+-K^+-ATP酶活性水平,改善肾功能衰竭大鼠脑组织的氧化应激。
Objective:To explore the effects of modified Huanglian Wendan Decoction on MDA,SOD and Na^+-K^+-ATPase in cerebral tissue of chronic renal failure (CRF) rats. Methods:Forty Wistar rats were randomly divided into blank control group,sham operation group,CRF model group and traditional Chinese medicine(TCM) group. Each group had 10 rats. The blank control group,sham operation group and CRF model group were given normal saline by gavage,while the TCM group was given modified Huanglian Wendan Decoction by gavage. After 6 weeks,the levels of MDA,SOD and Na^+-K^+-ATPase activity in cerebral tissue were tested. Results:Compared with the blank control group and the sham operation group,the MDA level in the CRF model group and the TCM group was significantly increased(P〈0.05);compared with the CRF model group,the MDA level in the TCM group was significantly reduced(P〈0.05). Compared with the blank control group and the sham operation group,the SOD level in the CRF model group was significantly reduced (P〈 0.05 ) , and there was no significant difference in the TCM group. Compared with the CRF model group,the SOD level in the TCM group was significantly higher(P〈0.05). Compared with the blank control group and the sham operation group,the Na^+-K^+-ATPase activity in the CRF model group and the TCM group was significantly lower(P〈0.05). Compared with the CRF model group,the Na^+-K^+-ATPase activity in the TCM group was significantly higher(P〈0.05). Conclusion :Modified Huanglian Wendan Decoction can significantly reduce MDA level,increase SOD level and Na^+-K^+-ATPase activity in cerebral tissue of 5/6 nephrectomy rat and ameliorate oxidative stress in brain tissue of CRF rats.
