四川部分地区肉牛鼻腔中支原体的PCR检测及分析 被引量:2

PCR detection and analysis of mycoplasma in the nasal cavity of beef in part of Sichuan province

摘要 为了解四川部分地区肉牛呼吸道病例中支原体的带菌情况及种类,从四川部分地区6个牛场的54头有呼吸道症状的肉牛鼻腔采取的棉拭子,经支原体属特异性PCR方法检测,并对特异性PCR产物克隆测序后比对分析鉴定支原体的种类;用牛支原体、牛眼支原体、殊异支原体和牛鼻支原体的特异性检测引物对这54份样本进行了检测,调查支原体感染情况.结果从54份肉牛鼻腔棉拭子样本中检出23份支原体阳性样本;鉴牛支原体(12/54)、牛眼支原体(6/54)、殊异支原体(3/54)和牛鼻支原体(2/54)共4种支原体.4种支原体特异性检测结果显示,牛支原体的感染率为57.3%,殊异支原体的感染率为29.63%,牛眼支原体检出率为20.37%,牛鼻支原体的检出率为16.67%.在两种支原体混合感染中,检出率相对较高的是以牛支原体+殊异支原体模式的混合感染,检出率为16.67%;三种支原体混合感染的情况有出现,而四种支原体混合感染的情况没有被检出.结果表明,四川省肉牛呼吸道疾病样本中支原体感染较为普遍,且种类较多,其中牛支原体检出率最高,首次在国内牛中检出牛眼支原体,研究结果丰富了国内牛呼吸道疾病的病原谱. In order to investigate the mycoplasma in beef nasal cavity in Sichuan province, firstly, a specific PCR method for detecting mycoplasma was used to test 54 nasal cavity swabs collected from beeves suffering from respiratory disease in 6 cattle farms in Sichuan province, and then, the PCR products were cloned and sequenced to identify mycoplasma species. The samples were tested to investigate the infection of mycoplasma using the specific primers for Mycoplasma bovis ,Mycoplasma bovoculi ,Mycoplasma clispar, and Mycoplasma bovirhinis. The results showed that total 23 mycoplasma positive samples were checked out from 54 samples including 4 species of mycoplasma such as Mycoplasma bovis (12/54),Mycoplasma bovoculi (6/54),Mycoplasma dispar (3/54) and Mycoplasma bovirhinis (2/54). The results of the four species of mycoplasma showed that the infection rates of Mycoplasma boris were 57.3%. The infection rates of Mycoplasma dispar were 29.63% ,the detection rate of Mycoplasma bovoculi were 20. 37% , and the detection rates of Mycoplasma bovirhinis were 16. 67%. Among the two mycoplasma mixed infections, the relatively high detection rate was a mixed infection of Mycoplasma boris and Mycoplasma dispar. The detec- tion rates were 16. 67% and the mixed infection of three mycoplasma cases appeared, but the mixed infection of four mycoplasma cases was not in beef samples. The results indicated that mycoplasma was common, and at least 4 species of mycoplasmas existed in beef respiratory in Sichuan province, among which,Mycoplasma boris was the most common. Meanwhile, this is the first report about the detection of Mycoplasma bovoculi. This report enriched the pathogenic Mycoplasma spectrum in domestic beef respiratory diseases.
