异丙酚对人肝癌细胞侵袭转移能力及上皮间质转化的影响 被引量:4

摘要 目的探讨异丙酚对人肝癌细胞侵袭转移能力及上皮间质转化(EMT)发生的作用。方法用逐渐增量(5、10、20μg/m L)的异丙酚刺激肝癌细胞(HCCLM3、QGY-7703、SMMC-7721)后,通过体外的细胞功能学实验(划痕实验、Transwell、三维培养实验)检测异丙酚对肝癌细胞迁移和侵袭能力的影响。20μg/m L的异丙酚刺激肝癌细胞,Western blot与免疫荧光实验检测刺激后HCCLM3细胞EMT相关标志物(Ecadherin、Vimentin)的表达情况。结果随着异丙酚浓度的提高,划痕实验显示肝癌细胞的迁移能力下降,Transwell侵袭实验和三维培养实验显示肝癌细胞侵袭能力下降(P<0.05)。20μg/m L异丙酚刺激HCCLM3细胞后,肝癌细胞的EMT得到抑制,上皮标志物E-cadherin表达上调,而间质标志物Vimentin表达下调(P<0.05)。结论在体外细胞模型中,异丙酚5~20μg/m L呈剂量依赖性抑制肝癌细胞的转移与侵袭,机制可能与其抑制EMT的发生有关。 Objective To investigate the influence of propofol on invasion,metastasis and epithelial-mesenchymal transition(EMT) of human hepatocellular carcinoma cells. Methods Hepatocellular carcinoma cells(HCCLM3,QGY-7703,SMMC-7721) were stimulated with propofol in gradually increasing dosages(5 μg/m L,10 μg/m L and 20μg/m L). The effects of propofol on the migration and invasion of hepatocellular carcinoma cells were assessed by wound healing assay,trans-well matrix invasion assay and three-dimensional spheroid invasion assay. HCCLM3 cells were stimulated with propofol of 20 μg/m L. Western blot and immunofluorescence assay were used to detect the expression of EMT makers(E-cadherin and Vimentin). Results With the increase of propofol concentration,wound healing assay showed that the migration ability of hepatocellular carcinoma cells significantly decreased(P〈 0. 05); trans-well invasion experiments and three-dimensional culture experiments showed that the invasive ability of hepatocellular carcinoma cells significantly decreased(P 〈0. 05). The expression of E-cadherin was up-regulated and the expression of vimentin was down-regulated in HCCLM3 cells after propofol stimulation of 20 μg/m L(P 〈0. 05). Conclusion In vitro,propofol can suppress the migration and invasion of HCC cells with concentration-dependent effect,which may be correlated to the inhibition of EMT.
