莪棱胶囊对子宫内膜异位症模型大鼠VEGF、bFGF及PDGF表达的影响 被引量:11

摘要 目的观察莪棱胶囊对子宫内膜异位症大鼠模型的治疗作用及血管内皮生成因子(VEGF)、碱性成纤维细胞生长因子(bFGF)和血小板衍生生长因子(PDGF)的影响,探讨莪棱胶囊抑制子宫内膜异位症相关血管生成的作用机制。方法取雌性未孕SD大鼠,除正常组12只大鼠外,采用自体移植法建立大鼠子宫内膜异位症模型,造模成功的大鼠随机分为正常组、模型组及药物高、中、低剂量组(4、2、1g/kg),每组12只。连续灌胃给药4周后,观察异位病灶体积与HE病理学变化;ELISA与HIC法检测VEGF、bFGF和PDGF的血清含量和组织表达量。结果药物组异位病灶体积大小明显降低(P<0.05),病理组织形态有改善;与正常组比较,模型组血清VEGF含量及子宫内膜异位组织VEGF、bFGF、PDGF表达明显升高(P<0.01),血清bFGF、PDGF含量无明显变化(P>0.05);与模型组相比,药物组血清及组织中VEGF、bFGF、PDGF的水平显著降低(P<0.05,P<0.01)。结论莪棱胶囊能明显抑制大鼠子宫内膜异位组织发展,其机制可能与抑制异位病灶血管生成相关。 Objective To study the treatment effect of ELeng Capsule in rat models of endometriosis,to investigate its effect on vascular endothelial growth factor(VEGF),basic fibroblast growth factor(b FGF) and platelet derived growth factor(PDGF),and to discuss its mechanism of angiogenesis inhibition. Methods Female SD rats were selected. Endometriosis model was constructed with auto grafting method. The endometriosis rats were randomly divided into model group,ELeng Capsule high-dose group,medium-dose group and low-dose group. After treatment for 4 weeks,the lesion tissues were collected. The levels of VEGF,b FGF and PDGF were detected by immunohistochemistry and enzyme linked immunosorbent assay(ELISA). Results Eleng Capsule significantly inhibited the endometriosis of rats in drug groups(P 〈0. 05),and alleviated pathological change in lesions. Compared with the normal group,the expression of VEGF in serum and the levels of VEGF,b FGF and PDGF in endometriosis tissues of rats in model group was significantly higher(P 〈0. 01),but there was no significant difference in the expression of b FGF and PDGF between model group and normal group(P〉 0. 05). Compared with model group,the levels of VEGF,b FGF and PDGF in blood serum and endometriosis tissues of rats in drug groups were significantly lower than model group(P〈 0. 05). Conclusion ELeng Capsule inhibits the growth of endometriosis tissues in rats. ELeng Capsule significantly suppresses endometriosis angiogenesis in rat model and it may have a closed relation with the inhibition of angiogenesis.
