摘要
目的探讨miR-124及STAT3在骨肉瘤组织中的表达,以及miR-124对骨肉瘤MG-63细胞增殖和侵袭的影响。方法 Real-time PCR法检测67例患者骨肉瘤组织及对应的癌旁组织中miR-124的表达,免疫组织化学方法检测STAT3蛋白的表达。使用脂质体将miR-124抑制剂及miR-124模拟物转染至骨肉瘤MG-63细胞株,以无关序列作为阴性对照。Western blot方法检测转染后STAT3蛋白的表达变化,采用MTT方法与Transwell实验检测转染后MG-63细胞的增殖侵袭能力变化。结果 miR-124在骨肉瘤组织中的表达明显低于癌旁组织(P<0.01);STAT3蛋白在骨肉瘤组织中的表达明显高于癌旁组织(P<0.01);转染miR-124抑制剂的MG-63细胞增殖侵袭能力与对照组相比显著升高,细胞内STAT3蛋白表达亦明显升高;转染miR-124模拟物后则得到相反的结果(P<0.01)。结论 miR-124抑制骨肉瘤MG-63细胞的增殖和侵袭可能与下调STAT3的表达相关。
Objective To investigate the expression of miR-124 and signal transducer and transcription activator 3(STAT3) in osteosarcoma tissue and the effect of miR-124 on the proliferation and invasion of MG-63 cells. Methods The expression of miR-124 in 67 cases of osteosarcoma and corresponding adjacent tissues was detected by Real-time PCR, and the expression of STAT3 protein was detected by immunohistochemistry. Mi R-124 inhibitors and miR-124 mimics were transfected into osteosarcoma MG-63 cell lines using liposomes, and the unrelated sequences were used as negative controls. Western blot method was used to detect the expression of STAT3 protein after transfection. The proliferation and invasion ability of MG-63 cells after transfection were detected by MTT and Transwell assay respectively. Results The expression level of miR-124 in osteosarcoma was significantly lower than that in adjacent tissues, the expression level of STAT3 protein was significantly higher in osteosarcoma tissues than in adjacent tissues(P〈0.01). Compared with the control group, the proliferation and invasion of MG-63 cells as well as the expression level of STAT3 was significantly increased after miR-124 inhibitor was transfected; the reverse results were obtained after miR-124 mimics was transfected(P〈0.01). Conclusion The inhibition of miR-124 on the proliferation and invasion of MG-63 cells might be related to downregulating of STAT3 expression.
