“调气”电针远端腧穴对CSR大鼠局部脊髓组织中JAK1、STAT3蛋白表达的影响 被引量:9

Effect of Electroacupuncture at Distal Acupoints on Expressions of JAK1 and STAT3 Proteins in Local Spinal Cord Tissue of CSR Rats

摘要 目的:以JAK1、STST3蛋白为指标探讨"调气"电针远端腧穴对神经根型颈椎病大鼠的镇痛机制。方法:将SPF级Wistar大鼠60只雌雄各半,随机分为正常组,模型对照组,"调气"电针组。除正常组外均造成神经根型颈椎病模型,"调气"电针组造模后第3天开始电针治疗,参照大鼠穴位图谱取双侧合谷、太冲,每天1次,每次20 min,连续治疗7 d后处死。模型对照组于造模后第3天开始,每日抓取1次,不做针刺处置。正常组不做任何处置。治疗后对3组大鼠采用YLS-3E型痛分析仪测量机械性痛阈,采用SABC免疫组织化学染色法对各组造模段脊髓及神经根组织,分别进行IgG染色,进行JAK1、STAT3免疫组化测定。结果:(1)造模后模型组各大鼠的痛阈值较正常组明显下降(P〈0.05);治疗后电针组各大鼠的痛阈值较模型组明显升高(P〈0.05);(2)与正常组相比,模型组的脊髓段及神经根组织中JAK1蛋白、STAT3蛋白表达显著升高,差异具有统计学意义(P〈0.05);与模型组相比,"调气"电针组的表达显著降低,差异具有统计学意义(P〈0.05)。结论:"调气"电针远端腧穴对CSR大鼠具有镇痛作用,对CSR脊髓及神经组织的JAK1-STAT3信号转导通路的抑制可能是实现其镇痛作用的重要机制之一。 Objective: To investigate the analgesic mechanism of Tiaoqi electroacupuncture(EA) at the distal acupoints on cervical spondylotic radiculopathy in rats with JAK1 and STST3 proteins as indicators. Methods: Sixty male and female rats of SPF grade Wistar rats were randomly divided into normal group,model control group and Tiaoqi EA group.In addition to the normal group,the other groups were set up cervical spondylotic radiculopathy type model. After modeling,on 3rd day,Tiaoqi EA group began EA treatment,by the reference acupoints map of rats,selecting bilateral Hegu and Taichong,once a day,20 min at a time. After continuous treatment for 7 days,the rats were put to death. The model control group began to crawl once a day on the third day after modeling,without acupuncture treatment. Normal group did not do any treatment. After treatment,three groups' rats were treated with YLS-3 E pain analyzer to measure the mechanical pain threshold. SABC immunohistochemical staining was used to stain each model's spinal cord and nerve root tissues. IgG staining was performed and JAK1 and STAT3 immunohistochemistry were performed. Results:(1)After modeling,the pain threshold of the rats in the model group was significantly lower than that in the normal group(P〈0. 05).After treatment,the pain threshold value of EA group was significantly higher than that of model group(P〈0. 05).(2)Compared with the normal group,the expressions of JAK1 protein and STAT3 protein in the spinal cord and nerve root tissue of the model group were significantly increased and the difference was statistically significant(P〈0. 05). Compared with the model group,the expression of EA group was significantly lower and the difference was statistically significant(P〈0. 05). Conclusion: Tiaoqi EA at the distal acupoints has an analgesic effect on CSR rats. The inhibition of JAK1-STAT3 signal transduction pathway in spinal cord and nerve tissue of CSR may be one of the important mechanisms to achieve its analgesic ef
