摘要
目的:探究冰片促进左旋多巴吸收进入血液和通过血脑屏障进入脑组织的机制及不同剂量的冰片对左旋多巴促进作用的差异性。方法:采用大鼠灌胃给药10d,解剖取血清和脑组织,用6%高氯酸沉淀蛋白后,采用HPLC法测定不同组别血清和脑浆中左旋多巴的含量,色谱条件为:Kromasil 100—5C18色谱柱(250mm×4.6mm E76863),以乙腈:0.1%三氟乙酸6:94为流动相进行洗脱.柱温35℃,流速1.0mL·min^-1,检测波长280nm。结果:与左旋多巴组比较,血清和脑浆样本中,冰片高剂量组和冰片中剂量组中左旋多巴的含量明显增加,差异有统计学意义(P〈0.05)。冰片低剂量组与左旋多巴组比较,左旋多巴的含量有所增加。但差异无统计学意义(P〉0.05)。结论:冰片在给药量0.0143g·kg^-1~0.1430g·kg^-1范围内能够促进左旋多巴吸收进入血液和脑组织.且在这一浓度范围内冰片高剂量组能更多地促进左旋多巴进入血清和脑组织,这可能与冰片降低大鼠小肠和脑组织中P-糖蛋白的含量有关。
Objective:To explore whether Bingpian(Borneolum syntheticum) can promote levodopa absorption into the bloodstream and can promote levodopa through the blood-brain barrier into the brain tissue and to explore whether there are differences of the different doses of borneol promoting the effects of levodopa. Methods:The medicine was given to the rats by lavage for 10 days. The serum and brain tissues were ob- tained after anatomy. After using 6% perchloric acid protein deposition,the contents of levodopa in serum and brain tissues of different groups were determined by HPLC. The chromatographic conditions including: Kromasil 100-5C18 chromatographic column (250 mm×4.6 mm E76863 ), acetonitrile:0.1% trifluoroacetic acid 6:94 as mobile phase elution. Column temperature was 35℃,flow rate 1.0 mL·min^-l,the detection wave-length was 280 nm. Results:Compared with the levodopa group,in the serum and brain samples,the contents of levodopa in the high dose borneol group and the medium dose borneol group were significantly increased (P〈0.05),the difference was statistically significant. Compared with the levodopa group,the content of lev-odopa was increased in the low dose group(P〉0.05),without statistical significance. Conclusions:The amount of borneol in the range of 0.0143--0.1430 g·kg^-1 can promote the absorption of levodopa into the blood and brain tissue. In the concentration range of Bingpian(Borneolum syntheticum),high dose group can promote more levodopa absorption into the blood and brain tissue,which might be related to borneol reducing the content of P-glycoprotein in the small intestine and brain tissue of the rats.
