喷雾干燥法固化姜黄素纳米混悬剂工艺及体外溶出度研究 被引量:5

摘要 目的提高姜黄素(curcumin,CUR)的溶出度。方法采用湿法研磨技术制备姜黄素纳米混悬剂(curcumin nanosuspension,CUR-NS),采用喷雾干燥技术固化CUR-NS。结果制得的CUR-NS为均匀的黄色混悬液,平均粒径为288 nm,跨度为0.702。喷雾干燥得到的CUR-NS喷干粉(curcumin spray dried powder,CUR-SDP)能够保持CUR-NS的粒度分布。CUR-SDP显著提高了CUR的体外溶出度。在20 min时CUR-SDP完全溶出,而CUR原料药(curcumin bulk drug,CUR-Bulk)和物理混合物(curcumin physical mixtures,CUR-PM)的累积溶出度只有49.2%和52.7%。结论通过湿法研磨技术降低药物粒径,能够显著提高水难溶性药物CUR的体外溶出度。 Objective To improve the dissolution of curcumin( CUR). Methods Curcumin nanosuspension( CUR-NS) was prepared by wet grinding technology,and CUR-NS was solidified by spray drying technique. Results The resulting CUR-NS was homogeneous yellowsuspension with a mean particle size of288 nm and the span of 0. 702. The CUR-NS spray dried powder( CUR-SDP) obtained by spray drying could maintain the particle size distribution of CUR-NS. CUR-SDP significantly increased the in vitro dissolution of CUR. CUR-SDP was completely dissoluted within 20 min,while the accumulated dissolution of CUR bulk drug( CUR-Bulk) and the physical mixture( CUR-PM) was only 49. 2% and 52. 7%. Conclusions The in vitro dissolution of poorly water soluble drug CUR can be significantly improved by reducing the particle size of the drug through wet grinding technology.
