摘要
【目的】鉴定在广东、海南和云南三省橡胶树苗圃病害调查时发现的引发橡胶树叶枯新病害——毛色二孢叶斑病的致病菌。【方法】采用组织分离法对从苗圃采集到的8份橡胶叶褐斑病病叶进行分离,获得10株病原菌菌株。通过形态学、生理生化、分子系统学(r DNA-ITS和EF1-α基因的PCR扩增、序列分析和系统进化树的构建)和致病性测定对病原菌进行了鉴定。【结果】该病原菌的菌落特征、显微形态特征和生理生化特性与可可毛色二孢菌(Lasiodiplodia theobromae)相一致,且该菌与其他作物可可毛色二胞菌株聚在系统发育树的1个分支上。【结论】经鉴定该橡胶树叶斑病的病原菌为可可毛色二孢菌(L.theobromae),这是首次在中国橡胶树上发现由该病菌引起的病害。
[Purpose] In order to classify the causal pathogen of a new disease of leaf spot on rubber tree was found at rubber tree nurseries in Guangdong, Hainan and Yunnan Province during a survey of rubber tree disease, causing necrosis and discolor of leaves and stems or even dead back.[ Method] Ten isolates were obtained from eight samples of diseased rubber tree leaves through a method of tissue isolation and classified by morphological observation, physiology and biochemistry study, sequence analyses of r DNA-ITS and EF1-α, and its pathogenicity was confirmed by Koch 's postulate.[ Result] The colony characteristics, microscopic morphological characteristics and physiological and biochemical characteristics of the pathogen were consistent with that of Lasiodiplodia theobromae. [Conclusion]The pathogen was identified as L. theobromae. The fungal species is firstly reported as the pathogen of rubber tree leaf spot.
