摘要
目的探讨犀角地黄汤的作用机制。方法从多个中药成分数据库、反向分子对接平台、蛋白数据库、疾病数据库中获取犀角地黄汤的化学成分、作用靶点、相关疾病,进而构建化合物-靶点网络、靶点-疾病网络、蛋白互作网络(protein-protein interaction,PPI),并对靶点蛋白进行基因本体中生物学过程(GO-BP,gene ontology-biology process)分析、基因组百科全书(Kyoto encyclopedia of genes andgenomes,KEGG)通路分析,从而来探讨其作用机制。结果经过筛选后获得71个候选化合物,237个相应蛋白靶点、201种相关疾病,hub靶点10个,GO-BP富集分析得条目160条,涉及信号转导、转录翻译、物质代谢、细胞凋亡和增殖、免疫等生物过程。KEGG分析得信号通路101条,涉及肿瘤、炎症、内分泌、肝病、代谢等相关通路。结论本研究初步分析了犀角地黄汤的作用及其机制,为进一步临床研究及新药开发提供了思路及基础。
Objective To study the efficacy and the mechanism of Xijiao Dihuang Tang. Methods Using traditionalChinese medicine databases, protein databases, disease databases and reverse molecular docking platform to getchemical components, targets and diseases related to four herbs of Xijiao Dihuang Tang. The compound-targetnetwork, the target-disease network, protein-protein interaction(PPI) network were established and the geneontology-biology process(GO-BP) analysis of target, the Kyoto encyclopedia of genes and genomes(KEGG)pathway enrichment analysis of targets were conducted to explore the mechanism of Xijiao Dihuang Tang. Results Totally 71 candidate compounds,237 targets,201 diseases,10 hub targets were obtained after screening. In GO-BP enrichment analysis,there were 160 terms involving signal transduction,transcriptional translocation,metabolicphase, apoptosis and proliferation, immune process. The KEGG pathway enrichment analysis showed 101 pathways,including pathways involved in cancer,inflammation,endocrine,liver disease,metabolic,and so on.Conclusion The preliminarily results of the study on the effects and mechanisms of Xijiao Dihuang Tang providedbases and novel ideas for further clinical research and new drug development.
