摘要
为研究酶法制备具有免疫调节作用的青蛤多肽,以青蛤肉为原料,分别采用胰蛋白酶、碱性蛋白酶、胃蛋白酶、木瓜蛋白酶和中性蛋白酶进行酶解,以小鼠巨噬细胞RAW 264.7的相对增殖率为指标,筛选出最佳酶种,再通过单因素试验和L_(16)(4~5)正交试验确定该酶的最佳酶解条件。结果表明:最佳酶种是胃蛋白酶,最佳酶解条件为p H2、温度42℃、料液比1︰10(g/m L)、加酶量1 600 U/g、时间8 h,所得酶解产物对RAW 264.7细胞的相对增殖率为76.15%。该工艺研究为青蛤免疫调节多肽的分离纯化及开发具有免疫调节作用的功能食品提供试验依据。
The optimal technology of immunomodulatory peptides prepared from Cyclina sinensis by enzymatic hydrolysis was studied.Use the Cyclina sinensis meat as raw material,choose the best enzyme from trypsin,alkaline protease,pepsin,neutral protease and papain by the relative proliferation rate of RAW264.7 macrophages.The optimum enzymatic conditions were determined by single factor experiment and L(16)(4^5) orthogonal experiment.The results showed that the optimal protease was pepsin and the optimum conditions of enzymatic hydrolysis were found to be p H of 2,temperature of 42 ℃,material liquid ratio of 1︰10(g/m L),the amount of enzyme of 1 600 U/g and enzymolysis time 8 h,the relative proliferation rate of RAW 264.7 macrophages of protein hydrolysates was 76.15% under optimum enzymatic conditions.These results suggested that protein hydrolysate from Cyclina sinensis might have applications as functional foods with immunomodulatory effects.
