摘要
目的:以川贝母鳞茎外表基部为材料,研究激素、暗处理时间与低温处理温度对鳞茎诱导效果的影响,为川贝母组培技术应用研究提供参考。方法:通过采用组织培养的方法,比较无菌处理的外植体在不同培养方式中的生长情况,并简要分析川贝母组织培养的影响因素。结果:低温暗处理,GA3均有利于打破鳞茎休眠,加快分化速度,同时GA3有提高愈伤组织诱导率的效果;这批试验样本中,愈伤组织诱导率最高为80.2%,不定芽诱导率最高为79.3%。结论:诱导生成不定芽的最佳激素配比为6-BA 2 mg·L^(-1)+NAA 0.5 mg·L^(-1),诱导鳞茎生成愈伤组织的最佳激素配比为6-BA1 mg·L^(-1)+NAA 2 mg·L^(-1)+GA3 1 mg·L^(-1);低温暗处理诱导鳞茎分化效果最佳的组合为4℃+5 d。
Objective:With Chuanbeimu bulb appearance base as the materials,the effects of hormone,dark processing and low temperature on the bulb was investigated to provide a reference for the tissue culture technology of Chuanbeimu.Method:Using tissue culture method,the growth conditions of explants in different culture medium were compared and the influence factors of Chuanbeimu tissue culture was analyzed.Result:Low temperature,dark environment and GA3 were conductive to break the dormancy of the bulb and accelerate the differentiation rate.GA3 also improved the frequency of callus induction.In these samples,the highest frequency of callus induction was 80.2%,and the highest rate of adventitious buds inductivity was 79.3%.Conclusion:The best hormone combination for inducing buds from bulb is 6-BA 2 mg·L^-1 + NAA 0.5 mg·L^-1.The best hormone combination for inducing callus formation from bulb is 6-BA 1 mg·L^-1 + NAA 2 mg·L^-1 + GA3 1 mg·L^-1.The optimum combination of low temperature and dark environment for inducing bulb is 4 ℃ +5 d.
