摘要
目的:建立常州市固定血小板捐献者人类白细胞抗原(HLA)的资料库,了解现有资料库中HLAA、B、DRB1位点分布的多态性。方法:收集常州市中心血站血小板捐献者标本125份,采用特异引物-聚合酶链反应(PCR-SSP)的方法检测HLA-A、B、DR基因,对基因分型的结果进行统计。结果:该血小板库HLA-A位点检出11个位点;HLA-B位点检出23个位点;HLA-DRB1位点检出13个位点。结论:现有的资料库中HLA-A、B、DRB1位点具有多态性,可为多次输注血小板产生HLA抗体的患者提供无HLA抗体相应抗原的单采血小板,以期取得较好的疗效。
Objective:To establish a database of human leukocyte antigen(HLA)in fixed platelet donors of Changzhou to understand the polymorphism of HLA-A,B,DRB1 loci in existing databases.Method:A total of 125 samples from Changzhou fixed platelet donors were genotyped for HLA by PCR-SSP and the genotype frequency.Result:11 loci were detected in the HLA-A locus of the platelet bank.23 loci were detected in the HLA-B locus and13 loci at the HLA-DRB1 locus.Conclusion:The HLA-A,B,DRB1 loci in the existing database are polymorphic and can provide single-shot platelets without HLA antibody antigens for patients with HLA-PAs.
