二甲双胍通过AMPK信号通路对小鼠骨关节炎保护作用探讨

摘要 目的:分析二甲双胍通过AMPK信号通路对小鼠骨关节炎保护作用。方法:选择C57BL/6健康小鼠40只(体重24~26g),随机分成对照及观察组,每组各20只,对照组不做任何处理(普食),观察组小鼠利用切除前交叉韧带和半月手术诱导膝关节不稳原理建立OA模型,成功建模后每天二甲双胍灌胃,对照组生理盐水灌胃,测量2组小鼠实验前、建模成功、灌胃60天膝关节直径,血TNF-α、HA及IgM;获取2组小鼠膝关节软骨,行体外实验培养24小时,观察AMPK信号表达情况。结果:观察组小鼠建模成功膝关节直径均高于实验前、灌胃60天及对照组(P〈0.05);观察组小鼠建模成功血TNF-α、HA水平均显著高于实验前、灌胃60天及对照组(P〈0.05);2组实验前后血IgM水平差异均不大(P〉0.05);膝关节软骨经体外实验培养后,其AMPK表达情况比较明显。结论:二甲双胍通过AMPK信号通路,提高局部组织氧供应,改善局部骨细胞能量代谢,进而对骨关节炎病灶产生一定的保护作用。 Objective: To investigate the protective effect of metformin on osteoarthritis in mice through AMPK signaling pathway. Methods: Forty healthy mice C57BL/6 (weighing 24 -26 g) were randomly divided into control and observation groups,20 in each group. The control group without any treatment ( general diet), in the observation group, the OA model was established by using the anteri-or cruciate ligament and semilunar surgery to induce the instability of the knee joint. The mett0rmin was given into the stomach atter suc- cessful modeling, and the control group was given normal saline gavage, then two groups of mice were measured before the experiment, the established model, and the diameter of the knee, blood TNF-a, HA and IgM were observed after 60 days. Then two groups of mice were obtained from the articular cartilage. The cells were cultured in vitro for 24 hours, and the expression of AMPK signal was observed. Re- suits: The diameters of successful knees of model mice in the observation group were higher than those before the experiment ,60 days after the operation and the control group ( P 〈 0.05). The levels of TNF - u and HA in the model blood of the mice in the observation group were significantly higher than those before the experiment, as well as those in the control group ( P 〈 O. 05). The blood IgM levels before and after the two groups were not significantly different( P 〉 0.05 ) ;After cultured in vitro,the expression of AMPK in knee cartilage was more obvious. Conclusion: The metformin could increase the local tissue oxygen supply through the AMPK signaling pathway,which im- prove the energy metabolism of local bone cells, and exert some protective effect on the osteoarthritis lesions.
