摘要
目的:建立花生衣中儿茶素、原花青素B2、表儿茶素和表儿茶素没食子酸酯的含量测定方法。方法:采用Kromasil C_(18)色谱柱(250×4.6 mm,5μm);流动相:乙腈-0.3%磷酸梯度洗脱;流速:0.7 mL/min;柱温:30℃;进样量:10μL;检测波长278 nm。结果:儿茶素、原花青素B2、表儿茶素和表儿茶素没食子酸酯分别在0.155~1.162μg、0.156~1.168μg、0.161~1.204μg和0.151~1.128μg范围内线性关系良好,回归方程分别为Y=5.367×10~5X+1.231×10~4,r=0.999 7;Y=5.639×10~5X+1.475×10~4,r=0.999 5;Y=6.151×10~5X+4.007×10~3,r=0.999 6;Y=4.874×10~5X+3.277×10~4,r=0.999 6。平均加样回收率分别为98.3%(RSD=1.2%)、97.6%(RSD=1.7%)、98.0%(RSD=1.0%)和98.2%(RSD=1.3%)。结论:本研究建立的方法简便、高效、快速、准确性高、重复性好,可用于花生衣的质量控制。
Objective:To establish determination methods of the content of catechin, procyanidin B2, epicatechin and epicatechin gallate in peanut skin. Methods:Adopted the Kromasil C18 chromatographic column(250×4.6 mm, 5 μm). The mobile phase was acetonitrile-0.3% phosphoric acid with gradient elution. The flow velocity was 0.7 mL/min. The column temperature was 30℃. The injection volume was 10 μL. The detection wavelength was 278 nm. Results: The catechin,procyanidin B2, epicatechin and epicatechin gallate respectively had good linearity in the ranges of 0.155 - 1.162 μg,0.156-1.168 μg,0.161-1.204 μg and 0.151-1.128 μg. The regression equation respectively was Y=5.367×10^5 X+1.231×10^4,r=0.999 7;Y=5.639×10^5 X+1.475×10^4,r=0.999 5;Y=6.151×10^5 X+4.007×10^3,r=0.999 6;Y=4.874×10^5 X+3.277×10^4,r=0.999 6. The average recovery respectively was 98.3%(RSD=1.2%),97.6%(RSD=1.7%),98.0%(RSD=1.0%),and 98.2%(RSD=1.3%). Conclusion:The method established in the study is simple,efficient and rapid with high accuracy and good reproducibility,which can be used for the quality control of peanut skin.
