摘要
探讨如何提高生物样品电镜图像的反差。通过改良传统染色方法、控制样本体积、把握染液pH值及温度、改进水洗工具、喷涂碳离子、碳膜捞片、减少切片厚度、选择小孔径光阑、降低加速电压、选择正确的欠焦等多方面因素,分析增加图像反差的方法。相同倍率下,采用提高电镜图像反差的办法能够得到更为清晰的超微结构。只有注意到每一个实验操作环节,才能得到较好的图像反差效果。
To explore the effective way to improve the contrast of electron microscopic images of biological samples, a lot of methods, such as adjusting the traditional dyeing method, controlling the sample volume, regulating the pH value and temperature of the dye solution, improving the water washing tool, spraying carbon ions, using the carbon film fillet, reducing the slice thickness, selecting the smaller aperture, lowering the acceleration voltage, selecting the correct under focus and were tested to increase contrast ratio of the image. It was found that the improving methods could get a clearer uhrastructure with the same magnification, and only each experimental operation is noted, then we can get a better effect of the contrast image.
