摘要
为探讨Lnc01089与肝癌细胞侵袭的关系,本研究分析了Lnc01089在肝癌细胞株和肝癌组织中的表达情况。从肝癌细胞株HepG2、Huh7、SMMC-7721和15例成对的肝癌组织及其癌旁组织中提取总的RNA,经逆转录得到cDNA,通过qRT-PCR实验,分析Lnc01089在肝癌组织和这三种肝癌细胞株中的表达水平。结果发现Huh7、SMMC-7721中Lnc01089的表达量明显低于HepG2;肝癌组织中Lnc01089的平均表达量也明显低于癌旁组织,差异均有统计学意义(p〈0.05)。结果提示Lnc01089表达水平的降低与肝细胞的恶性程度有关。因此Lnc01089可能与肝癌的发生、发展有密切联系,有望为肝癌治疗提供新的靶点。
In order to investigate the relationship between Lnc01089 and the invasion of liver cancer cells, we analyzed the expression of Lnc01089 in liver cancer cell lines and liver cancer tissues. Total RNA was extracted from Hep G2, Huh7, SMMC-7721 hepatoma cell lines and 15 cases paired of liver cancer tissues as well as adjacent non-tumor tissue. Then c DNA was obtained by reverse transcription, and the expression level of Lnc01089 in HCC tissues and three HCC cell lines was analyzed through q RT-PCR. The results showed that the expression of Lnc01089 in Huh7 and SMMC-7721 cell lines was significantly lower than that of Hep G2 cell line,and the average expression of Lnc01089 in hepatocellular carcinoma tissues was also significantly lower than that of the normal adjacent tissues, the difference was statistically significant(p〈0.05). The results indicated that the decrease of Lnc01089 expression level might be related to the malignant degree of liver cells. Therefore, Lnc01089 may be closely related to the occurrence and development of HCC, which might provide a new target for the treatment of liver cancer.
