蟾酥干燥炮制前后化学成分和药效学变化考察 被引量:14

Study on chemical constituents and pharmacodynamic changes of Bufonis Venenum with different drying methods

摘要 目的考察蟾酥Bufonis venenum鲜浆干燥炮制加工(日晒干燥、50℃真空干燥、50℃鼓风干燥、80℃鼓风干燥、冷冻干燥)前后化学成分及药效变化。方法 HPLC法和TLC法研究蟾酥不同干燥方式炮制前后化学成分的变化,MTT法测试不同炮制干燥品抑制5种不同肿瘤细胞株增殖的效果。结果 5种不同加工炮制方法所得蟾酥,性状有较大差异;化学成分的种类及含量没有明显差异;华蟾酥毒基和脂蟾毒配基含量之和都符合《中国药典》2015年版标准,大于6%;50℃和80℃鼓风干燥所得蟾酥具有比其他干燥方法更强的肿瘤细胞抑制效果。结论日晒及50℃鼓风干燥法所得蟾酥符合《中国药典》要求的外观性状。真空干燥和冷冻干燥虽然颜色不符合《中国药典》的规定,但主要有效成分并未发生较大变化。 Objective To study the changes of chemical constituents and pharmacodynamics with different drying methods(sundrying, 50 ℃ vacuum-drying, 50 ℃ and 80 ℃ heat-drying, freeze drying method) in Bufonis Venenum. Methods HPLC method and TLC method was established for studying the changes of chemical constituents from B. Venenum before and after being dried, and determine the inhibitory effect of dried samples on five different tumor cell lines proliferation by MTT assay. Results The B. Venenum processed by five different methods were different in character, while no significant differences in the types and content of chemical constituents; The total content of resibufogenin and cinobufagin was more than 6%, which was consistent with the 2015 edition of Chinese Pharmacopeia; 50 ℃ and 80 ℃ heat-drying of B. Venenum showed more effective inhibitiory effect than other dry methods. Conclusion The appearance of B. Venenum met the criterion of pharmacoperia by sun-drying and 50 ℃ heat-drying method. Although the color of B. Venenum under the vacuum-drying and freeze drying method did not meet the requirements of Chinese Pharmacopeia, the main effective components have a few changes.
