摘要
目的观察益气活血解毒方对A549/DDP耐药细胞JAK2-STAT3信号通路分子JAK2、STAT3表达的影响。方法培养A549细胞、A549/DDP细胞,分别用高、中、低剂量益气活血解毒方含药血清进行干预,MTT法筛选最佳浓度的含药血清,流式细胞术检测细胞凋亡率,Western blot技术检测各组细胞中JAK2、STAT3蛋白表达变化,RT PCR技术检测各组细胞中JAK2、STAT3 mRNA表达变化。结果益气活血解毒方含药血清作用48h能明显抑制A549/DDP细胞增殖,且最佳浓度为中剂量;与对照血清组比较,益气活血解毒方最佳含药血清组A549/DDP细胞凋亡率明显提高(P<0.05);与A549细胞组比较,A549/DDP细胞组JAK2、STAT3蛋白及mRNA表达明显降低(P<0.05),与对照血清组比较,益气活血解毒方最佳含药血清组A549/DDP细胞JAK2、STAT3蛋白及mRNA表达明显升高(P<0.05)。结论益气活血解毒方干预肺癌化疗耐药的作用可能是通过调节JAK2-STAT3信号通路,促进细胞凋亡实现的。
Objective： To observe the effect of Yiqi Huoxue Jiedu Decoction on the expression changes of JAK2-STAT3 signaling pathway of A549/DDP resistant cells. Methods： The A549 and A549/DDP ceils were cultured and intervened with high,medium and low doses of Yiqi Huoxue Jiedu Decoction,and the best concen- tration of drug-containing serum of Yiqi Huoxue Jiedu Decoction was Screened by MTr;the cell apoptosis rate was detected by flow cytometry,the protein and mRNA expression of JAK2 and STAT3were detected by Western- blot and RT PCR. Results： The drug-containing serum of Yiqi Huoxue Jiedu Decoction could obviously inhibit the proliferation of A549/DDP cells after 48 hours,and the best concentration was medium dose group ;compared with the blank serum group,the apoptosis rate of A549/DDP ceils of medium dose group increased significantly （P〈 0.05 ）;compared with A549 group ,the protein and mRNA expression of JAK2 and STAT3 of A549/DDP cell group decreased significantly（P〈 0.05）,while compared with the blank serum group,the JAK2 and STAT3 protein and mRNA expression of medium dose group of Yiqi Huoxue Jiedu Decoction increased significantly （P〈 0.05）. Conclusion： The mechanism of Yiqi Huoxue Jiedu Decoction on interventing chemotherapeutic resistance of lung cancer may be related to promoting cell apoptosis by regulating PI3K and AKT protein expression.
