PTEV干扰对甲状腺滤泡上皮细胞Nthy-ori3-1侵袭能力的影响及其相关机制研究

摘要 目的研究PTEN干扰对甲状腺滤泡上皮细胞Nthy—ori3—1侵袭能力的影响,并初步探讨其可能的分子机制。方法以人正常甲状腺滤泡上皮细胞Nthy—ori3—1为研究对象,通过慢病毒感染技术将PTEN特异性短发夹RNA(shRNA)及无关序列shRNA导入细胞。实验分为2组:无关序列组(shContr01)和干扰PTEN组(shPTEN)。通过Transwell小室侵袭实验检测细胞的侵袭能力,免疫荧光检测侵袭相关蛋白在细胞表面的表达水平,Westernblot检测相关蛋白的表达水平。结果与无关序列组相比,干扰PTEN组细胞的形态发生明显改变,侵袭相关蛋白E—cadherin的表达水平明显降低,细胞的侵袭能力增强,侵袭相关蛋白E—cadherin在细胞表面的表达水平降低。结论PTEN干扰能够促进甲状腺滤泡上皮细胞Nthy—ori3—1的侵袭,且这一作用可能是通过下调侵袭相关蛋白E—cadherin的表达实现的。 Objective To investigate the effects of PTEN interference on the invasive ability of thyroid follicular epi- thelial cells and to explore its possible molecular mechanism. Methods A human normal thyroid follicular epithelial cell line ( Nthy - ori3 - 1 ) was used, to which PTEN - targeted short hairpin RNA (shRNA) and unrelated sequence - targe- ted shRNA were transfected by lentivirus. There were two groups in the experiments: an unrelated sequence (shControl) group and a PTEN interference (shPTEN) group. The invasion ability of ceils was detected by Transwell cell invasion as- say. The level of invasion related proteins were detected by immunofluorescence. The level of related proteins were detec- ted by Western blotting. Results Compared with the shControl group, the shPTEN group showed remarkable morpholog- ical changes and marked enhanced invasive abilities, with significantly reduced levels of E - cadherin, especially on the cell surface. Conclusions PTEN interference can promote the invasion of Nthy - ori3 - 1 cells, which may be mediated by down - regulating expression of E - cadherin.
