不同剂量骨碎补总黄酮对大鼠Masquelet技术诱导膜中BMP-2和VEGF表达的影响 被引量:11

Effects of Different Dose of Rhizoma Drynariae on Expression of BMP-2 and VEGF in Membrane of the Rats Induced by Masquelet Technology

摘要 目的:观察不同剂量的骨碎补总黄酮对大鼠Masquelet技术诱导膜中骨形成蛋白-2(Bone Morphogenetic Protein,BMP-2)和血管内皮生长因子(Vascular Endothelial Growth Factor,VEGF)表达的影响。方法:选取48只3月龄SD大鼠,雌雄不限,按随机数字表法分为4组:空白对照组、低剂量组、中剂量组及高剂量组,每组12只。所有大鼠均建立左侧胫骨长段骨缺损模型,并予以抗生素骨水泥填塞。术后低、中、高剂量组分别给予骨碎补总黄酮67.5,202.5和607.5mg/(kg·d)灌胃,空白对照组给予等量蒸馏水灌胃,连续灌胃6周后取出骨水泥,切取诱导膜,检测诱导膜中BMP-2和VEGF的表达情况。结果:灌胃6周后,各组BMP-2和VEGF表达组间比较,差异有统计学意义(P<0.05);各组两两比较,低、中、高剂量组BMP-2和VEGF表达均显著高于空白对照组,且高剂量组表达最明显,中剂量组次之,低剂量组再次之,呈剂量依赖关系,差异均有统计学意义(P<0.05)。结论:不同剂量的骨碎补总黄酮均可促进大鼠Masquelet技术诱导膜中BMP-2和VEGF的表达,且呈剂量依赖关系,高剂量组最明显。 Objective:To observe the effects of different dose of rhizoma drynariae on the expression of bone morphogenetic protein(BMP-2)and vascular endothelial growth factor(VEGF)in membrane of rat which induced by masquelet technology.Methods:Forty-eight SD rats without limitation of sex were randomly divided into four groups:control group,low dose group,middle dose group and high dose group.All rats were established with long tibial long bone defect model,and antibiotic bone cement was filled.The low,medium and high dose groups were given 67.5,202.5 and 607.5 mg/(kg·d)of the total flavonoids.The expression of BMP-2 and VEGF in the induced membrane and the growth of the blood vessel were measured after 6 weeks of continuous administration.Results:After 6 weeks of intragastric administration,the expression of BMP-2 and VEGF had compared each other,the difference were statistically significant(P〈0.05).All groups multiple comparison,the expression of BMP-2 and VEGF in the low,middle and high dose groups was significantly higher than that in the blank control group.And the highest expression was in the high dose group,the second was in the middle dose group,and the third was in the low dose group.There was a dose-dependent relationship,and the differences were statistically significant(P〈0.05).Conclusion:Different doses of total flavonoids of rhizoma drynariae can promote the expression of BMP-2 and VEGF in membrane of rat which induced by masquelet technology in a dose-dependent manner.
