摘要
目的:观察骨碎补总黄酮对体外培养大鼠成骨细胞血管内皮生长因子(vascular endothelial growth factor,VEGF)和成纤维细胞生长因子2(fibroblast growth factor 2,FGF-2)表达以及碱性磷酸酶(alkaline phosphatase,ALP)活性和矿化结节的影响。方法:在大鼠成骨细胞体系中加入不同浓度的骨碎补总黄酮作用24 h和48 h。采用ELISA和RT-PCR法检测VEGF和FGF-2表达变化,PNPP法检测ALP活性以及von Kossa染色检测矿化结节形成。结果:骨碎补总黄酮可刺激VEGF和FGF-2,使其在蛋白和mRNA水平表达均明显升高,显著提高成骨细胞ALP的活性和促进矿化结节的形成。结论:骨碎补总黄酮可提高成骨细胞VEGF和FGF-2的表达,促进其成骨作用。
Objective: To observe the effect of Drynaria flavonoids on the expression of vascular endothelial growth factor ( VEGF )and fibroblast growth factor 2 ( FGF-2 )and alkaline phosphatase ( ALP ) activity and the formation of mineralized nodules on the rat osteoblast. Methods : Rat osteoblasts were treated by different concentrations of Drynaria flavonoids for 24- h and 48 h. The expressions of VEGF and FGF-2 were detected by ELISA and RT-PCR. ALP activity was determined by PNPP assay and the formation of mineralized nodules was observed by von Kossa staining. Resuhs: Drynaria flavonoids can significantly promote the VEGF and FGF-2 expressions at the protein and mRNA levels, the ALP activity and the formation of mineralized nodules on rat osteoblasts. Conclusion : Drynaria total flavonoids can promote the VEGF and FGF-2 expressions and improve the osteogenesis.
