摘要
本文采用正交设计试验,以挥发油包合率和包合物收率为考核指标,优选挥发油β-CD包合工艺,采用单因素试验法,以颗粒剂的制粒情况为考核指标,优选颗粒剂的最佳制备工艺.确定挥发油β-CD包合物最佳制备工艺和颗粒剂的最佳制备工艺.发现最佳包合工艺条件为:采用胶体磨法制备挥发油β-CD包合物,β-CD与挥发油的比例为8∶1,加10倍量蒸馏水,研磨30 min;颗粒剂最佳制备工艺为:药物粉末与可溶性淀粉和糊精的比例为9∶0.5∶0.5,以80%乙醇为润湿剂,制软材,制颗粒,于50℃下干燥,整粒,即得.从而确定了包合工艺与颗粒剂的制备工艺设计合理、结果稳定,适合于大工业生产.
Objective: To optimize the inclusion technology of volatile oil by β-CD and its forming process. Methods: With the inclusion rate of volatile oil and inclusion compound yield as assessment index,the inclusion technology of volatile oil was optimized by orthogonal experimental design; to select the forming process using wet granulating technology by single factor experiment. Results: The optimized inclusion conditions was β-CD and volatile oil mixed by 8∶ 1,10-fold water was added into the mixture and the mixture was triturated for 30 min by colloid mill; the best forming process was: drug powder、amylum solubila and microcrystalline cellulose mixed by 9 ∶ 1 ∶ 1,the granule was prepared by wet granulating technology with 80% ethanol,and dry at 50℃. Conclution: The optimized inclusion technology and forming process was reliable、fundamental steady and the feasible. It is comfortable for big factory production.
