速溶性聚合物微针的制备及性能研究 被引量:4

Study on the preparation and performance of rapidly dissolving polymer microneedles

摘要 目的 以聚乙烯吡咯烷酮(PVP)为基质,制备适用于水溶性或醇溶性药物给药的速溶性微针,并研究其性能。方法分别采用离体乳猪皮进行微针皮肤穿刺试验,采用白色家兔进行体内溶解试验,并进行皮肤刺激性试验及体外透皮试验,比较微针给药和溶液给药的透皮特性。结果 所制备的微针能刺入离体猪皮而不折断,机械性能良好;刺入家兔皮肤5 min后,针体基本可完全溶解;空白微针刺入皮肤时表现出轻刺激性;以钙黄绿素为模型药物,微针组乳猪皮的经皮渗透速率为0.0278μg·cm^-2·h^-1,是溶液组的28倍。结论 所制备的微针性能优良,可用于水溶性或醇溶性药物的给药。 OBJECTIVE To prepare rapidly dissolving polymer microneedles fitting for water - soluble or ethanol - soluble drugs with PVP and to investigate their performance. METHODS Insertion experiment using neonatal porcine skin in vitro was performed. The kinetics of dissolution was characterized by inserting microneedles into rabbit skin. The dermal irritation experiment was carried out. At last, Drug transdermal characteristics of mieroneedles were compared with solution' s by transdermal experiment in vitro. RESULTS The polymer microneedles could be inserted into neonatal porcine skin without breaking off, showing good mechanical properties. Remaining 5 min after insertion in skin, the microneedles could be dissolved. The blank microneedles could cause light irrita- tion to rabbit skin. In transdermal delivery experiment, using calcein as a model drug, the penetration rate of microneedles group was 0. 0278μg·cm^-2·h^-1 ,which was 28 times of the solution group. CONCLUSION The polymer microneedles showed good property, and could be used for loading of water - soluble or ethanol - soluble drugs.
