西洋参多糖提取与纯化工艺优化 被引量:3

Optimization of extraction and purification of polysaccharide from Panax quinquefolium L.

摘要 目的:筛选西洋参多糖的最佳提取及纯化工艺.方法:以西洋参多糖为指标,采用正交实验对料液比、煎煮时间与次数进行优化.以吸附率为指标,对上样液质量浓度、上样液pH、上样流速、径高比进行正交实验优选;以解吸率为指标,对乙醇体积分数、洗脱剂pH、洗脱流速采用正交实验进行优化.结果:料液比、提取次数是西洋参多糖提取的关键影响因素,西洋参多糖最佳提取工艺为料液比1:10,加水提取3次,每次1.5h.上样液质量浓度、上样液pH、上样流速、径高比为多糖纯化的关键影响因素,多糖的纯化工艺为上样质量浓度2.74 mg/mL,上样液pH 8.0,上样流速2.0 mL/min,径高比1:8,最大上样量3.0 mL,洗脱剂为体积分数30%乙醇,洗脱剂pH 8.0,洗脱流速1.0 mL/min,洗脱剂用量3.0 mL;得到西洋参多糖纯度为47.94%.结论:建立的方法稳定、可靠,为开发利用西洋参资源提供基础. Objective:This paper aimed to optimize the extraction techniques and purification process of polysaccharide from Panax quinquefolium L. Methods: Conditions for extraction was screened by the content of polysaccharide, and conditions for purification was screened by the adsorption rate and the desorption rate, as described by orthogonal experiment design. Results: Solid to liquid ratio and cooking time had notable influence on the extraction of polysaccharide. The optimum extraction conditions for polysaccharide from Panax quinquefolium L: Solid to liquid ratio 1: 10, extraction for 3 times with 1.5 h for each. Concentration of the upper sample liquid, sample pH, sample flow rate, and diameter to heightratio had notable influence on the purification of polysaccharide. The optimum purification conditions for polysaccharide : sample concentration 2.74 mg/mL, sample pH 8.0, flow rate 2.0 mL/min, diameter to height ratio 1: 8, maximum sample size 3 mL, desorption agent 30% alcohol, desorption agent pH 8.0, flow rate 1.0 mL/min and desorption agent 3.0 mL; the purity of polysaccharide from Panax quinquefolium L. was 47.94%. Conclusion: Through the present study on the extraction and purification technology for polysaccharide from Panax quinquefolium L., we demonstrated that the optimal extraction and purification processes were stable and reliable, which may provide a methodology for developing and utilizing the resources of Panax quinquefolium L..
