摘要
目的优化油菜籽中总酚提取工艺,建立油菜籽总酚测定方法,评估不同产地油菜籽中总酚含量差异。方法以芥子酸为对照品,采用Folin-Ciocaileu比色法测定油菜籽中总酚含量,检测波长725 nm。同时探究了甲醇浓度、提取时间、料液比对油菜籽中总酚提取的影响,确定了最佳提取工艺,并测定了我国7个省份不同产地油菜籽总酚含量。结果实验确定了最佳提取工艺:料液比为1:20(g/mL),提取溶剂为50%甲醇,提取时间为3 h。芥子酸的线性回归方程为Y=0.1487X+0.0137,相关系数r^2=0.9996,线性范围为0.5~4μg/mL。油菜籽总酚含量范围为4455.21~7842.30 mg/kg。结论 Folin-Ciocaileu比色法简单、快速、准确,是油菜籽中多酚含量测定的可靠方法。不同产地油菜籽中总酚含量存在显著性差异,其中贵州省油菜籽总酚含量最高。
Objective To optimize the extraction process of total phenol in rapeseed, establish a method for determination of total phenol content in rapeseed, and evaluate the differences of total phenol content in rapeseed from different producing regions. Methods Taking sinapinic acid as reference, the content of total phenol in rapeseed was determined by Folin-Ciocaileu colorimetry at detection wavelength of 725 nm. The effects of methanol concentration, extraction time and solid-liquid ratio were investigated, respectively. The total phenol content in rapeseed from 7 provinces were determined and compared. Results The optimal extraction conditions were as follows: the solid-liquid ratio was 1:20 (g/mL), the extraction solvent was 50% methanol and the extraction time was 3 h. The linear regression equation of sinapinic acid was Y=0.1487X+0.0137, while the correlation coefficient r2 was 0.9996 and the linear range was from 0.5 to 4 μg/mL. The total phenolic content in rapeseed samples ranged from 4455.21 to 7842.30 mg/kg. Conclusion Folin-Ciocaileu colorimetry is simple, rapid and accurate, and it is suitable for the determination of total phenol content in rapeseed. Besides, there are significant differences in total phenol content of rapeseed from different producing areas, and the highest content of total phenol in rapeseed is from Guizhou province.
