摘要
目的探讨Runx3和c-Myc蛋白在骨肉瘤组织中的表达及其在MG-63细胞增殖过程中的作用。方法采用Western blot技术检测骨肉瘤组织和骨良性病变组织中Runx3和c-Myc蛋白表达水平;MTS方法检测TGF-β对骨肉瘤细胞系MG-63细胞增殖能力的影响。结果肉瘤组织中Runx3蛋白的表达水平明显低于骨良性病变组织,而c-Myc则明显高于骨良性病变组织(P<0.05),二者呈负相关(R=-0.797,P<0.05)。TGF-β抑制MG-63细胞增殖活性,并呈剂量依赖关系,TGF-β可显著增强MG-63细胞Runx3蛋白表达并抑制c-Myc蛋白表达;高表达Runx3蛋白可减弱TGF-β对MG-63细胞增殖的抑制作用(P<0.05)。结论TGF-β抑制MG-63细胞增殖可能与上调Runx3蛋白表达进一步抑制c-Myc蛋白的表达相关。
Objective To study the role of Runx3 and c-Myc proteins in the inhibitory effect of TGF-βon MG-63 cells proliferation.Methods Western blot was used to detect Runx3 and c-Myc protein expression in osteosarcoma and benign bone tissues.MTS method was used to determine the effect of TGF-βon the proliferation of osteosarcoma cell line MG-63.Results The expression level of Runx3 protein was significantly lower in osteosarcoma tissues than in benign bone lesions but higher for c-Myc protein(P〈0.05),with negative correlation(R=-0.797,P〈0.05)between Runx3 and c-Myc.TGF-βinhibited significantly MG-63 cell proliferation in a dose-dependent manner,but enhanced Runx3 expression and inhibited c-Myc protein expression in MG-63 cells(P〈0.05).However,the overexpression of Runx3 protein attenuated the inhibition effect of TGF-βon the proliferation of MG-63 cells(P〈0 05).Conclusion The inhibition effect of TGF-βon MG-63 cells proliferation might be related to upregulation of Runx3 protein and further inhibition of c-Myc expression.
