摘要
为利用大别山野生刺楸资源,基于响应面法优化了刺楸叶总皂苷的酶法提取工艺条件,并探讨其抗氧化活性。结果表明,最佳提取工艺条件为纤维素酶添加量1.1%,乙醇体积分数73%,料液比1∶15(g∶m L),酶解温度71℃,酶解时间2 h。在此条件下,刺楸叶总皂苷提取率为2.52%,分别为传统醇提和水提工艺提取率的1.5倍和2倍,所得酶提液质量浓度为0.37 g/L时与水提液的羟自由基抑制能力无显著差异,是相同质量浓度VC的1.78~4.45倍,表明酶法提取工艺适合产业化生产,酶提液具有较强的抗氧化活性,从而为大别山刺楸植物资源的综合利用提供了试验依据。
To utilize the wild Kalopanax septemlobus resource in Dabie Mountains, the enzymatic extraction process conditions of total saponins from K. septemlobus leaves were optimized based on response surface methodology. Moreover, the antioxidant activity of total saponins from K. septemlobus leaves was studied. The results showed that the optimum extraction conditions were cellulase addition 1.1%, ethanol concentration 73%, solidliquid ratio 1∶15(g∶ml), hydrolysis temperature 71 ℃ and time 2 h. Under the conditions, the extraction rate of total saponins from K. septemlobus leaves was 2.52%, which was 1.5 and 2 times of traditional alcohol extraction and water extraction process, respectively. The hydroxyl radical inhibiting ability between 0.37 g/L enzyme extracts and water extracts obtained had no significant difference, and they were 1.78-4.45 times that of the same mass concentration of VC, which revealed that the enzymatic extraction process was suitable for industrial production and the extraction liquid had strong antioxidant activity. This work provided the experimental basis for the comprehensive utilization of K. septemlobus resources in Dabie Mountains.
