摘要
目的:探讨胆管癌中Smad7基因单核苷酸多态性(SNP)及蛋白表达与胆管癌的相关性。方法:选择胆管癌490例用于Smad7单核苷酸多态性研究,胆道非肿瘤性病变887例作为对照。其中选取胆管癌140例,胆道非肿瘤性病变100例用免疫组化检测组织中Smad7蛋白的表达。Hi-SNP结合多重PCR技术和高通量测序技术检测单核苷酸多态性。结果:SNP结果显示SMAD7rs12953717型与胆管癌的发生风险显著正相关,而rs4939827型与胆管癌的发生风险显著负相关,二者间存在线性关系(P<0.01)。Smad7在胆管癌中的阳性表达率显著高于正常胆管上皮的阳性表达率。胆管癌中Smad7表达与淋巴结转移、浸润范围、神经侵犯、组织学分级有显著性相关(P<0.05)。Smad7阳性表达病例的总生存率比阴性病例更短。结论:Smad7 SNP与胆管癌的发生风险有关,且其蛋白表达与胆管癌的恶性演进和预后有关。
Objective: To investigate the association of single nucleotide polymorphism and protein expression of Smad7 gene with cholangiocarcinoma in cholangiocarcinoma. Methods:490 cases of choice bile duct cancer were studied by Smad7 single nucleotide polymorphism, 887 cases of non-tumorous biliary duct pathological change were acted as control comparison. Immunohistochemical technique was used to detect the expression of Smad7 protein in 140 cases of bile duct cancer and 100 cases non-tumorous biliary duct pathological change. Hi-SNP combined with multiplex PCR and high-throughput sequencing technology to detect single nucleotide polymorphism. Results: The SNP results showed that SMAD7 rs12953717 was significantly positively related to the risk of cholangiocarcinoma in this group, whereas rs4939827 was negatively relationship between them associated (P〈0.01) with the risk of cholangiocarcinoma, and there was a linear The positive expression rate of Smad7 in cholangioearcinoma was significantly higher than that in normal bile duct epithelium. The expression of Smad7 in bile duct carcinoma was significantly correlated with lymph node histological grade (P〈0.05). The overall survival rate negative cases. Conclusion: Smad7 SNP are related to metastasis, invasion extent, invasion of nerve and in Smad7 positive cases was shorter than in the the risk of cholangioearcinoma, and its protein expression is related to the malignant progression and prognosis of cholangiocarcinoma
