甘草对重型颅脑损伤大鼠血清炎性细胞因子及BCL-2、BAX、NGF表达的影响 被引量:5

Effect of radix glycyrrhizae on serum inflammatory cytokines and expressions of BCL-2, BAX and NGF in rats with severe craniocerebral injury

摘要 目的:通过观察重型颅脑损伤大鼠血清TNF-α、IL-6的变化及大鼠脑组织神经生长因子(NGF)、BCL-2、BAX的表达,探讨甘草的神经保护作用。方法:72只雄性SPF级SD大鼠按照随机数字表法分为正常组、假手术组、模型组、甘草组。除正常组、假手术组外,其余两组建立大鼠重型颅脑损伤模型。造模后6 h,甘草组予以甘草水提取物溶液0.85 g/kg灌胃,其余三组予以等量的生理盐水灌胃。在24 h、72 h、168 h三个时间段采用双抗夹心ELISA法检测血清TNF-α、IL-6,免疫组织化学技术检测脑组织NGF、BCL-2和BAX。结果:(1)模型组在重型颅脑损伤后24 h即出现TNF-α、IL-6升高,72 h达到高峰,168 h有所回落,但仍高于正常组(P<0.05);甘草组TNF-α、IL-6含量在三个时间段均明显低于模型组(P<0.05)。(2)模型组NGF、BCL-2表达在24 h即明显增加,一直持续至168 h,但在三个时间段的表达差异无统计学意义(P>0.05);与模型组相比,甘草组NGF、BCL-2表达在24 h明显增加,在168 h达到高峰(P>0.05),呈现出用药时间的依赖性;(3)与正常组相比,模型组BAX表达在24 h即明显增加,72 h达到高峰,168 h稍有回落,仍高于正常组(P<0.05);甘草组BAX表达在三个时间段较模型组均明显降低(P<0.05)。结论:甘草能减轻重型颅脑损伤大鼠继发性炎症反应及细胞凋亡,具有神经保护作用。 Objective: Through observing the changes of serum TNF-ot,IL-6 and the expressions of brain tissue nerve growth facto(NGF) ,BCL-2 and BAX in rats with severe craniocerebral injury,to investigate the neuroprotec- tive effect of radix glycyrrhizae. Methods :72 male SD rats of SPF were randomly divided into normal group, sham operation group, model group and radix glycyrrhizae group. Except the normal group and sham operation group, the other two groups were established into the models of rats with severe craniocerebral injury. After 6h of modelling, the radix glycyrrhizae group was administrated 0. 85 g/kg aqueous extract of radix glycyrrhiza by gavage,while the other three groups were given the same amount of normal saline by gavage. In three time periods of 24h, 72h and 168h, TNF-α and IL-6 in serum were detected by double antigens sandwich ELISA,and NGF, BCL-2 and BAX in brain tissue were detected by immunohistochemistry. Results: 1. At 24h after severe craniocerebral injury, the model groupg content of serum TNF - α and IL-6 increased immediately, reached a peak at 72h, and fell back somewhat at 168h, but was still higher than normal group (P 〈 0. 05 ). TNF-α and IL -6 content of radix glycyr- rhizae group was obviously lower than model group in three time periods (P 〈 0.05 ). 2. The expression of NGF and BCL-2 in model group increased immediately obviously at 24h, until 168h, but the difference of expressions in three time periods was statistically sig- nificant ( P 〈 0. 05 ). Compared with model group, the expression of NGF and BCL-2 of radix glycyrrhizae group increased obviously at 24h, reached a peak at 168h, and showed the dependence on medicationtime.
