摘要
目的探讨地黄饮子对急性脑中风大鼠脑组织脑源性神经营养因子(BDNF)和基质细胞衍生因子1(SDF1)表达的影响。方法选取大鼠60只,随机分为正常组、治疗组和缺血组,各20只。麻醉后钳夹颈总动脉40 min,建立急性脑中风模型。治疗组:灌服地黄饮子汤36 g/kg,缺血组:灌服盐水1次,正常组:正常饮食。3周后,测量大鼠脑梗死面积,测定三组海马CA1区BDNF和SDF1蛋白表达情况。结果正常组脑梗死面积为0。治疗组脑梗死面积明显低于缺血组,P<0.05,差异有统计学意义。正常组海马CA1区BDNF和SDF1蛋白含量均明显高于治疗组和缺血组,P<0.05,差异有统计学意义。治疗组BDNF和SDF1蛋白含量明显高于缺血组,P<0.05,差异有统计学意义。结论地黄饮子通过提高BDNF和SDF1蛋白水平,增加内源性保护机制和神经干细胞增殖、迁移,保护神经元的功能。
Objective To explore the effect of Dihuang drink on the expression of BDNF and SDF1 in rats with acute cerebral apoplexy.Methods 60 rats were randomly divided into two groups:normal group,treatment group and ischemic group.After anesthesia,the carotid artery was 40 min and the model of acute cerebral apoplexy was established.Treatment group:potion of decoction of Dihuang drink(36 g/kg),ischemia group:1 time,normal group:normal diet.Three weeks later,the area of cerebral infarction was measured,and the expression of BDNF and SDF1 protein in three groups of hippocampal CA1 regions were measured.Results The normal area of cerebral infarction were 0.The area of cerebral infarction in the treatment group was significantly lower than the ischemic group P〈0.05,and the difference was statistically significant.The contents of BDNF and SDF1 protein in the normal group of hippocampus CA1 region were significantly higher than those in the treatment group and the ischemic group,P〈0.05 was statistically significant.The content of BDNF and SDF1 protein in the treatment group was significantly higher than that in the ischemic group P〈0.05,and the difference was statistically significant.Conclusion In order to protect the function of neuronal cells,it increases the level of BDNF and SDF1 protein and increases the proliferation and migration of endogenous protective mechanisms and neural stem cells.
