摘要
以西瓜的花粉为材料,研究西瓜花粉自发荧光的光谱特征、荧光强度,并结合结构照明显微技术优化花粉自发荧光成像方法。结果表明:西瓜花粉的自发荧光在蓝色波段最强;新鲜花粉在储藏过程中荧光强度变化不大;西瓜花粉的自发荧光主要位于花粉外壁网纹上;分别用苯胺蓝、Calcofluor white、碘化丙啶、吖啶橙对西瓜花粉染色后,花粉的其他部位在不同荧光通道下也可以呈现荧光。因此,本研究建立的方法不仅可用于分析西瓜花粉的大小、形态、外壁纹理、萌发孔、萌发沟等形态特征,还可以反映花粉表面荧光物质分布的特征。
With watermelon pollen as the material, the spectrum and intensity of the autofluorescence were investigated, and the photography of the autofluorescence of pollens were optimized combined with Apotome microscope. The results showed that the maximum intensity of autofluorescence was in blue spectrum; The intensity of autofluorescence in fresh pollens fluctuated only slightly during storage; And the autofluorescence was located mainly on the reticula. After staining the pollens with aniline blue, calcofluor white, propidiumiodide, acridine orange respectively, the other parts could also be fluorescent in specific channels. Therefore, the optimized method could be not only suitable for observing the size, shape, reticulum, aperture and germ furrow, but also indicate the distribution of chemical composition.
