摘要
试验旨在研究日粮中添加不同水平的青贮桑叶对陕北白绒山羊脂肪组织(皮下脂肪、尾部脂肪、肾周脂肪)、肝脏、肌肉组织中PPARγ、ADIPOQ、aP2mRNA表达的影响。选取8~9月龄体重相近体况基本一致的陕北白绒山羊羯羊40只,随机分为4组,分别饲喂含青贮桑叶为0%(Ⅰ组),10%(Ⅱ组),15%(Ⅲ组),20%(IV组)的日粮,试验期70d,其中预试期10d,正试期60d;饲养试验结束后屠宰,取皮下脂肪、尾部脂肪、肾周脂肪、肝脏、背最长肌组织进行RT-PCR,检测上述基因mRNA表达量。结果如下,PPARγ、ADIPOQ、aP2 mRNA在陕北白绒山羊不同组织部位均有表达,PPARγ、aP2mRNA在皮下脂肪组织表达量显著高于所检测的其他组织(P<0.05),ADIPOQ mRNA在脂肪组织的表达显著高于肝脏和肌肉组织(P<0.05)。在皮下脂肪组织和肝脏,PPARγ、ADIPOQ、aP2mRNA的表达量I组显著高于其他组(P<0.05),不同添加水平的青贮桑叶对其他组织部位PPARγ、ADIPOQ、aP2mRNA的表达量的影响不一致。结果表明,饲喂青贮桑叶能显著影响陕北白绒山羊脂肪代谢相关基因mRNA的表达,青贮桑叶可以作为潜在的具有降脂功效的饲料补充料添加到山羊饲料中以调节脂肪沉积改善肉品质。
This experiment was conducted to study the effects of diets supplemented with ensiled mulberry leaves on the mRNA expression of Peroxisome proliferator-activated receptor-γ(PPARγ),Adiponectin(ADIPOQ),adipocyte fatty acid-binding protein(aP2)in the adipose tissues(subcutaneous,tail,perirenal),liver and muscle of Shaanbei white cashmere goats.Forty Shaanbei white cashmere goats were selected and randomly divided into 4 groups.Control group was fed the basal diet(group Ⅰ);The experimental one was the basal diet supplemented with 10% (group Ⅱ),15% (group Ⅲ),20% (group IV)of ensiled mulberry leaves(dry matter basis)with pre-feeding period of 10 d,the formal period of 60 d,the trial period 70 d.At the end of experience,it respectively and randomly selects three goats from each group to slaughter trial and collects samples for required determination indicator.The results are as follows:PPARγ,ADIPOQ,aP2 mRNA expressed in all tissues studied.PPARγmRNA highly expressed in subcutaneous adipose tissue than other tissues(P<0.05),the same as aP2.ADIPOQ mRNA highly expressed in adipose tissues than in the liver and muscle(P<0.05).In subcutaneous adipose tissue and liver,PPARγ,ADIPOQ,aP2 mRNA highly expressed in group I than other groups(P<0.05).In other tissues,different levels of diets supplemented with ensiled mulberry leaves have different effects on expression of PPARγ,ADIPOQ,aP2 mRNA on Shaanbei White cashmere goats.Theses results suggest that the effects of diets supplemented with ensiled mulberry on PPAR-γ,ADIPOQ,aP2 mRNA expression were significant,ensiled mulberry leaves can be used as a potential fat-decreasing diet to adjust fat accumulation and improve meat quality for goats.
