“一标多测”多指标综合加权评分法优选牛舌草有效成分的提取工艺 被引量:5

Optimization of Extraction Technology for Anchusa italica Retz.by Multi-index Comprehensive Evaluation Method with “One Standard and Multi-index Determination”

摘要 提出了改进的多指标综合加权评分法,并将该方法用于优选意大利牛舌草中多种有效成分的提取工艺.针对样品中的未知含量分析物,提出了"一标多测"的概念,以意大利牛舌草中的多组成提取为例,选取芦丁为参照即为"一标",以此获得了5种主要指标组分的相对含量,即为"多测".以相对含量作为综合评价指标,通过对主要影响因素,包括提取溶剂、浸泡时间、超声时间、料液比等4个因素进行了正交试验优选.由此获得最佳提取工艺为:采用80%甲醇提取,浸泡12 h,超声10 min,料液比为1∶50.结果表明,"一标多测"解决了样品中被测物真实含量未知的问题,多指标综合加权评分法实现了实际样品未知含量分析物提取条件的优选.二者结合简便可行,节省试剂,并经过优选意大利牛舌草中多种待测成分的提取工艺得到了证实."一标多测"与多指标综合加权评分法结合对其他中药材中有效成分提取具有借鉴意义. A modified multi-index comprehensive evaluation method is applied to optimize the extraction technology of multi-components in Anchusa italica Retz. The content of rutin in Anchusa italica Retz. is selected as reference, so the relative contents of five main component are quantified by"one standard and multi-index determination"proposed for unknown compounds in sample. With the relative content of the main components as comprehensive evaluation index, four main extraction factors, including extraction solvent, soaking time, ultrasonic time, ratio of material to liquid, are optimized by orthogonal test.Optimum extraction technology was obtained as the sample was soaked for 12 h with 80% methanol and the sample/methanol ratio of 1:50, followed by sonicating for 10 min. It indicated that multi-index comprehensive evaluation method was simple, feasible, and economic."One standard and multi-index determination"is used as a solution to solve the problem that the content of the multi-components is unknown.The combination of the two techniques are validated with the optimization of extraction technology of multicomponents in Anchusa italica Retz. and could be used for the extraction of the active components from the other traditional Chinese medicine samples.
