摘要
目的:研究Netrin-1、DCC基因在6株不同类型和来源的体外培养结直肠癌细胞mRNA及蛋白水平的表达差异。方法:CCK8检测6株不同来源结直肠肿瘤细胞的生长曲线,以正常肠上皮细胞NCM460为对照;Real-Time PCR和Western blot分别检测Netrin-1、DCC基因在6株结直肠癌细胞内的mRNA和蛋白水平的表达。结果:与对照组相比,结直肠肿瘤细胞株可见DCC、Netrin-1基因及蛋白的异常表达;DCC表达在6株结直肠肿瘤细胞株中均表达下调,其中SW620、LOVO、LS174T细胞株内DCC表达下调差异显著,有统计学意义(P<0.05);Netrin-1在6株结直肠肿瘤细胞株中均表达上调,其中SW620、LOVO、LS174T细胞株内Netrin-1表达上调差异显著,有统计学意义(P<0.05);6株结直肠肿瘤细胞株之间DCC、Netrin-1也存在差异表达。结论:DCC、Netrin-1在结直肠肿瘤内异常表达,与肿瘤的发展和转移相关,DCC和Netrin-1在同一细胞株内表达呈负相关,DCC蛋白的下调和失活可能与Netrin-1有关。
Objective: To study the differential expressions of Netrin-1 and DCC gene in 6 colorectal cancer cell lines. Methods: CCK-8 method was used to detect the growth curve in SW480,HCT116,HT-29,SW620,LOVO and LS-174 T cells,and NCM460 was used as a control. Real-Time PCR and Western blot were used to detect different expressions of Netrin-1,DCCs mRNA and protein level in 6 colorectal cancer cell lines. Results: Compared with the control group( NCM460),there was abnormal expression of DCC,Netrin-1 and protein in 6 colorectal cancer cell lines; the expression level of DCC was down-regulated and showed significance in SW620,LOVO and LS174 T cell lines( P〈0.05),the expression level of Netrin-1 was up-regulated and showed significance in SW620, LOVO and LS174 T cell lines( P〈0.05).Conclusion: The expressions of DCC and Netrin-1 were abnormal in 6 colorectal cancer cell lines; DCC and Netrin-1 is negatively associated with tumor development and metastasis; and there is a correlation between Netrin-1 control and DCC down-regulated.
