高温胁迫对2种杂交石斑鱼存活率及血清生化指标的影响 被引量:14

Influence of High Temperature Stress on Survival Rate and Serum Biochemical Indexes of 2 Epinephelus Hybrids

摘要 对25℃条件下养殖的体长(33.5±2.4)cm、体质量(584.15±10.35)g的云纹石斑鱼(Epinephelus moara♀)×鞍带石斑鱼(Epinephelus lanceolatus♂)F1(简称云龙杂交斑),以及体长(25.1±2.2)cm,体质量(375.41±7.35)g的斜带石斑鱼(Epinephelus coioides♀)×鞍带石斑鱼(Epinephelus lanceolatus♂)F1(简称青龙杂交斑),以1℃/d的速度逐渐升高水温,记录各温度条件下两种杂交石斑鱼的存活率,研究二杂交石斑鱼的半致死温度及25、29、33、36℃条件下的血清生化指标。结果表明,云龙杂交斑和青龙杂交斑的高温半致死温度分别为34.9℃和37.9℃。随温度的升高,青龙杂交斑血清中甘油三酯(TG)、葡萄糖(GLU)含量及超氧化物歧化酶(SOD)、碱性磷酸酶(AKP)活力呈上升趋势,各温度组与25℃组差异有统计学意义(P<0.01),肌酐(CREA)含量下降,总胆固醇(T-CHO)29、33℃组下降(P<0.01),36℃组升至初始水平,各组谷草转氨酶(GOT)、谷丙转氨酶(GPT)和过氧化氢酶(CAT)活力无明显变化(P>0.05)。随温度的升高,云龙杂交斑血清中GLU、T-CHO含量呈上升趋势,33℃组与25℃组差异有统计学意义(P<0.01),CREA、TG含量略微升高后开始降低(P<0.05),GOT、AKP和酸性磷酸酶(ACP)的活力较25℃时下降(P<0.05),血清GPT和CAT活力无明显变化(P>0.05)。 Hybrid of Epinephelus moara♀× E. lanceolatus♂(i.e. Yunlong: BL33.5 ± 2.4 cm and BW 584.15 ± 10.35 g), and hybrid of E. coioides♀× E. lanceolatus♂(i.e. Qinglong: BL 25.1 ± 2.2 cm and BW 375.41 ± 7.35 g) were selected to investigate their tolerance to high temperature cultured condition. The fishes were cultured at the initial water temperature of 25 ℃ with a daily increment of 1 ℃. During the high temperature culture period, the survival rates were monitored and the median lethal temperature(LT50) for the hybrids was estimated. In addition, the changes in several serum biochemical parameters were determined at 25, 29, 33 and 36 ℃. The results indicated that the LT50 of Yunlong and Qinglong were 34.9 and 37.9 ℃, respectively. For the Qinglong hybrid, serum concentration of triglycerides(TG) and glucose(GLU), the activities of superoxide dismutase(SOD) and alkaline phosphatase(AKP) showed a significant increase with increasing cultured temperatures(P〈0.01). In contrast, serum concentration of creatinine(CREA) and total cholesterol(T-CHO) decreased initially and then increased(P〈0.01). Other serum biochemical indices such as glutamic-oxaloacetic transaminase(GOT), glutamate pyruvate transaminase(GPT) and catalase(CAT) showed no significant changes(P〈0.05). For Yunlong hybrid, serum concentration of T-CHO and GLU elevated significantly(P〈0.01) with the increase of culture water temperature, while the serum CREA and TG levels increased initially and then decreased at the end(P〈0.05). The serum activities of GOT, ACP and AKP showed a descending trend(P〈0.05) whereas the changes in serum activities GPT and CAT were insignificant(P〈0.05).
