摘要
目的:评价红景天苷对糖尿病心肌病(DCM)大鼠的疗效。方法:采用高脂饮食联合腹腔注射链脲佐菌素方法诱导雄性糖尿病大鼠,继续高脂高糖饮食,筛选糖尿病心肌病模型大鼠。将造模成功大鼠随机分为DCM组,红景天苷低、中、高剂量组及卡托普利组,另设正常对照组给予常规饲料,按组别不同分别给予药物干预,8周后采血检测各组空腹血糖、胰岛素、糖化血红蛋白、B型钠尿肽(BNP)及血管紧张素-Ⅱ(Ang-Ⅱ)水平;超声心动图评价各组大鼠心的结构和功能,观察各组大鼠心肌组织病理变化。结果:与DCM组比较,红景天苷各剂量组大鼠空腹血糖及糖化血红蛋白水平显著降低。与DCM组比较,红景天苷各剂量组大鼠BNP、Ang-Ⅱ水平显著下降,且与药物浓度呈正相关。与DCM组比较,红景天苷各剂量组大鼠左室舒张末压、左室收缩末期内径及左室舒张末期内径下降,射血分数显著升高。与DCM组比较,红景天苷各剂量组大鼠心肌组织结构紊乱程度减轻。结论:红景天苷可以改善糖尿病心肌病大鼠的心功能。
Objective: To evaluate the effect of salidroside on experimental rats with diabetic cardiomyopathy (DCM). Methods: Male diabetic rats were induced by intraperitoneal injection of streptozotocin. The rats were then fed with high-fat and high-sugar diet so as to select the DCM rat model. The modeled rats were randomly divided into DCM group, salidroside low-dose group, salidroside middle-dose group, high-dose salidroside group and captopril group, and another rats in the normal control group were given conventional food. After 8 weeks, the fasting blood glucose, insulin, glycosylated hemoglobin, B type natriuretic peptide (BNP) and angiotensin Ⅱ (Ang-Ⅱ) levels were detected in each group. Echocardiography was used to evaluate the cardiac structure and function, and the pathological changes of myocardial tissue were observed. Results: Compared with the model group, the levels of BNP and Ang-Ⅱ changed significantly in the salidroside drug intervention groups, and it was related to drug dosage. The degree of heart function damage decreased in the salidroside drug intervention groups. Conclusion: Salidroside may improve cardiac function in rats with diabetic cardiomyopathy.
