摘要
基于双缩脲反应和紫外分光光度法,建立一种简便、快速测定虫草花酶解液中肽含量的方法。以样品与双缩脲试剂的比例、反应时间、反应温度为自变量,以吸光度为因变量,对检测方法进行优化,并进行了回收率试验和精密度试验。结果表明:最佳的反应条件为样品:双缩脲试剂(v/v)=1.5:1,室温下反应5 min。线性回归方程为y=0.0947x+0.0062(R2=0.9977),虫草花酶解前和酶解后的肽含量分别为(13.094±0.528)mg/m L和(17.318±0.914)mg/m L。回收率为(104.54±2.22)%,精密度试验的相对标准偏差(RSD)为2.37%。综上所述,该方法的准确度好、精密度高,适用于虫草花酶解液中肽含量的测定。
Based on biuret reaction and ultraviolet(UV) spectorphotometry, a simple and rapid determination method of the content of peptides in cordyceps flower hydrolysate was established. Conditions of the reaction were optimized by using proportion of sample to biuret, reaction temperature and reaction time as the independent variable and absorbance as the dependent variable, and recovery test and precision test were conducted. Results showed that the optimal conditions were as follow: sample:biuret(v/v)=1.5:1, room temperature and reaction time 5 min. The linear regression equation was: y=0.0947 x+0.0062(R2=0.9977), and the content of peptides in cordyceps flower before and after hydrolysis were(13.094±0.528)mg/m L and(17.318±0.914)mg/m L, respectively. Recovery rate was(104.54±2.22)% and relative standard deviation(RSD) of precision test was 2.37%. In conclusion, the method possessed well accuracy and high precision, which was suitable for determination of the content of peptides in cordyceps flower hydrolysate.
