摘要
从海带中分离得到的12株乳酸菌中筛选得到一株具有较强产胞外多糖能力的菌株YYS-06,该菌株在发酵培养基中经72 h发酵可产154.6 mg/L多糖,经生理生化特性检测实验和16S r RNA序列分析将菌株YYS-06鉴定为植物乳杆菌(Lactobacillus plantarum)。通过发酵条件优化正交实验确定菌株YYS-06最适发酵条件为:接种量1%、发酵温度34℃、发酵时间36 h,在此发酵条件下胞外多糖产量为269.3 mg/L。目前YYS-06菌株被中国典型培养物保藏中心(CCTCC)保藏,编号为CCTCC NO M2016512。
A strain of lactic acid bacteria with higher yield of exocellular polysaccharide(EPS) was screened from 12 strains which were isolated from Laminaria japonica. The yield of EPS from lactic acid bacterial determined was 154.6 mg/L. The strain was identified as Lactobacillus plantarum by using biochemical tests and sequence analysis of 16 S r RNA. The optimal fermentation conditions with highest yield of EPS by using orhogonal methods were as fellows: the MRS media was fermentating 36 hours at 34 ℃ for culture temperature with the amount of inoculation of 1%. Under these conditions, the yield of EPS by Lactobacillus plantarum YYS-06 was 269.3 mg/L. The Lactobacillus plantarum YYS-06 strain was preserved by the China Center for Type Culture Collection(CCTCC) and the number is CCTCC NO. M2016512.
